小鼠脾脏基质微环境对浆细胞样树突状细胞的功能调控作用

【摘要】: 浆细胞样树突状细胞(pDC)是一类在免疫应答与免疫耐受中发挥重要作用的免疫细胞,在接受病原刺激后迅速活化,分泌大量的Ⅰ型干扰素,增强或调控其他免疫细胞的功能,有效地连接了固有免疫和适应性免疫。本课题利用小鼠内皮型脾脏基质细胞系模拟体内外周淋巴器官内免疫应答的微环境,研究了基质微环境对pDC的功能调节及其相关机制。结果提示:1.脾脏基质微环境抑制病毒活化的pDC继续分泌Ⅰ型干扰素、IL-12、TNF-α和pDC抗原提呈功能,同时抑制了活化pDC对NK细胞的激活效应,使NK细胞分泌IFN-γ下调。对其机制的研究表明基质细胞分泌的TGF-β参与了基质微环境对活化pDC的功能调控;2.脾脏基质微环境中的静息pDC在接受病毒刺激时仍然迅速活化,分泌高水平Ⅰ型干扰素、IL-12和TNF-α,发挥独特的抗病毒作用。因此,基质微环境对静息和活化pDC的调控作用使机体在免疫防御的同时,维持了免疫自稳,为临床上自身免疫疾病、过敏、以及感染等治疗方面提供了新的思路。
[Abstract]:Plasma cell like dendritic cell (pDC) is a kind of immune cells that play an important role in immune response and tolerance. After being stimulated by pathogen, plasmacyte-like dendritic cells (pDC) are activated rapidly, secreting a large amount of interferon type I, and enhancing or regulating the function of other immune cells. It effectively connects innate immunity with adaptive immunity. In this study, the mouse endothelial splenic stromal cell line was used to simulate the immune response of peripheral lymphoid organs in vivo and in vivo, and the functional regulation of matrix microenvironment to pDC and its related mechanism were studied. The result suggested that 1: 1. The splenic matrix microenvironment inhibited the viral activation of pDC and continued to secrete IL-12TNF- 伪 and pDC antigen presenting function. It also inhibited the activation effect of activated pDC on NK cells and down-regulated the secretion of IFN- 纬 by NK cells. The study of its mechanism indicated that TGF- 尾 secreted by stromal cells was involved in the regulation of pDC activation by matrix microenvironment. Resting pDC in splenic matrix microenvironment was still activated rapidly and secreted high levels of interferon type I IL-12 and TNF- 伪, which played a unique antiviral role. Therefore, the effect of matrix microenvironment on resting and activating pDC makes the body maintain immune self-stabilization while immune defense, which provides a new idea for the treatment of autoimmune diseases, allergies and infections.
