乳酸杆菌对细胞肿瘤坏死因子-α分泌的影响

【摘要】: 背景:益生菌对健康的积极作用越来越受到人们的重视。乳酸杆菌作为目前临床应用最为广泛的益生菌之一,其在IBD、IBS、慢性腹泻等有肠道炎症基础的疾病治疗中的重大价值已经被许多高水平的临床研究证实。然而关于益生菌对肠道炎症作用机制方面的研究虽然开展多年,但是对此问题仍没有统一全面的认识。肠上皮细胞和肠粘膜固有层免疫细胞作为肠道屏障的重要组成均有可能参与益生菌对肠道炎症的调控,它们各自在肠道炎症中的作用已被一些研究部分证实,但缺乏有效的研究比较两者在益生菌调控肠道炎症方面的作用。细胞因子TNF-α几乎存在于所有的炎症反应当中,它的强大的促炎症作用能够激活炎症介质联级反应并动员炎症细胞。测定TNF-α是评价肠道炎症的理想指标。 目的:研究乳酸杆菌(Lactobacillus casei CICC6002)对肠上皮细胞(HT-29)和单核细胞(THP-1)TNF-α分泌的影响,并对照非致病性大肠埃希菌(Escherichia coli DH5α)对细胞TNF-α分泌的影响情况,探索益生菌的抗炎作用及其作用途径。 方法:将37℃有氧条件下,MRS、LB细菌培养基中处于对数生长期的L casei、E coli各自转种于RPMI培养液孵育48小时,以100 000g×1小时,,4℃超速离心收集含细菌分泌代谢产物的条件培养基(CM);培养肠上皮细胞(HT-29)和单核细胞(THP-1);以脂多糖(LPS)刺激HT-29细胞和THP-1细胞分泌TNF-α,加入细菌的CM,共孵育4小时。以ELISA的方法检测L casei CM、E coli CM对HT-29细胞、THP-1细胞TNF-α分泌的影响。数据进行t检验。 结果:LPS刺激,加或不加细菌CM[956.39±374.45,475.74±49.59,829.62±110.51pg/ml],均使THP-1细胞TNF-α的分泌比基础状态[100.96±48.95pg/ml]明显增加(P<0.05)。与单纯LPS刺激的THP-1细胞TNF-α分泌[956.39±374.45pg/ml]相比,加入L casei CM显著减少LPS刺激的THP-1细胞TNF-α分泌[475.74±49.59pg/ml](P<0.05);加入E coli CM,LPS刺激的THP-1细胞TNF-α分泌[829.62±110.51pg/ml]无明显变化(P>0.05)。不加任何刺激、单纯LPS刺激或者LPS与L casei CM联合刺激条件下均未能检出HT-29细胞TNF-α分泌。LPS与E coli CM联合刺激可使HT-29细胞产生TNF-α[10.94±6.32pg/ml],明显低于相同条件下THP-1细胞TNF-α分泌[829.62±110.51pg/ml](P<0.01)。 结论:益生菌干酪乳酸杆菌具有抗炎特性,对抑制肠道炎症意义重大。干酪乳酸杆菌抗炎特性通过减少单核细胞分泌TNF-α实现。肠上皮细胞没有参与干酪乳酸杆菌的抗炎过程。非致病性大肠埃希菌DH5α无抗炎特性。
[Abstract]:Background: people pay more and more attention to the positive effect of probiotics on health. Lactobacillus is one of the most widely used probiotics in clinic at present. Its important value in the treatment of IBD,IBS, chronic diarrhea and other diseases based on intestinal inflammation has been confirmed by many high levels of clinical research. However, although the studies on the mechanism of probiotics on intestinal inflammation have been carried out for many years, there is still no unified and comprehensive understanding of this problem. As an important component of intestinal barrier, both intestinal epithelial cells and intestinal mucosal lamina immune cells may be involved in the regulation of intestinal inflammation by probiotics. Their respective roles in intestinal inflammation have been partly confirmed by some studies. But there is a lack of effective research to compare the role of probiotics in the regulation of intestinal inflammation. Cytokine TNF- 伪 exists in almost all inflammatory responses, its powerful pro-inflammatory role can activate the inflammatory mediators of the chain reaction and mobilize inflammatory cells. Determination of TNF- 伪 is an ideal index for evaluating intestinal inflammation. Objective: to study the effect of Lactobacillus (Lactobacillus casei CICC6002 on the secretion of TNF- 伪 in intestinal epithelial cells (HT-29) and monocytes (THP-1), and to explore the anti-inflammatory effect of probiotics and its pathways by comparing the effect of non-pathogenic Escherichia coli (Escherichia coli DH5 伪 on the secretion of TNF- 伪.
