SHIV-XJ02170病毒感染中国恒河猴的实验研究

【摘要】:SHIV病毒艾滋病动物模型是目前国外艾滋病模型研究的趋势,被认为是比现阶段普遍应用的SIV模型针对性更强、更有意义的动物模型。为了尽早开展国内此项研究,,尤其是选定毒力较强的SHIV毒株,成功建立SHIV感染动物模型,在前期SIV/SAIDS模型研究的基础上,本研究以SHIV-XJ02170病毒(以SIVmac239病毒基因组和C型HIV-1病毒的env基因为主体构建的一株SHIV病毒)及前病毒质粒接种中国恒河猴,进行了包括临床表现、病毒学、免疫学等方面研究。为了更准确定量测定实验恒河猴病毒感染状态,建立了测定SIV/SHIV病毒RNA载量的SYBR Green Ⅰ荧光染料实时定量RT-PCR方法和测定SIV/SHIV前病毒DNA的SYBR Green Ⅰ实时定量PCR方法。同时为了进一步增强SHIV-XJ02170病毒的毒力,对其进行中国恒河猴体内传代,以期获得一株高毒力、高致病性的SHIV病毒。 本研究共选用了13只中国恒河猴,实验前经血清学和PCR证明无SIV,STLV-1,SRV/D和B病毒感染。实验在生物安全三级实验室中进行。第一代实验猴共4只,分两组。第一组2只经肌肉注射SHIV-XJ02170前病毒质粒DNA;第二组两只静脉注射SHIV-XJ02170病毒液。当第一代实验猴检出病毒达高峰期后,取10mL感染猴的全血,抗凝后静脉接种第二代4只实验猴。第二代实验猴检出病毒后再将其全血静脉接种第三代3只实验猴,连续传至第4代2只猴。每代实验猴均定期采集肝素和EDTA抗凝血液标本,分别进行病毒分离、PCR方法检测感染猴PBMC中前病毒DNA、RT-PCR方法检测血浆病毒RNA、CD4~+/CD8~+、血浆P27抗原等指标的测定,以监测SHIV-XJ02170病毒在恒河猴体内复制和传代情况。实验结果显示原代SHIV-XJ02170病毒和前病毒质粒均可感染中国恒河猴,但传代的实验猴病毒检出率降低,病毒血症持续时间明显缩短,且全部13只实验猴都未产生典型的艾滋病症状,只有二代猴A0408
