羊胎盘免疫调节因子的免疫促进作用及其机理研究

【摘要】: 根据药物对机体免疫功能的不同影响,免疫药理学家一般把免疫调节剂分为免疫增强剂和免疫抑制剂两大类。前者的效应特点是促进免疫功能,而后者则表现为对免疫应答的抑制。羊胎盘免疫调节因子是本室采用现代生物技术以山羊胎盘为原料制备的生物活性物质,初步活性分析发现其具有免疫增强活性。在分析总结目前国内外有关动物源性免疫活性肽免疫增强作用特点研究资料的基础上,本研究根据免疫增强剂药效学研究原则,系统地探讨羊胎盘免疫调节因子的免疫促进作用及其功能特点,并对其作用机理进行分析研究;进而,对进一步分离纯化的单一组分之一的羊胎盘肽 Ⅰ的免疫调节作用进行初步研究,为羊胎盘活性成分的深入研究和开发提供基础研究资料,为GPIF的申报提供临床前的研究资料,为GPIF的临床应用提供科学的实验依据,对GPIF功能组分的分离纯化和进一步的研究开发奠定基础。 本研究共分四部分,分别就GPIF对BALB/c小鼠T细胞免疫功能、体液免疫功能和巨噬细胞免疫功能的促进作用进行系统研究,对GPIF免疫促进作用机理作部分探讨。在此基础上,对GPIF分离纯化的单组分之一GPIF Ⅰ的T细胞免疫功能进行初步研究。 第一部分,从体内、外两方面,建立环磷酰胺所致免疫抑制模型、~(60)Coγ射线辐射所致免疫抑制模型,探讨羊胎盘免疫调节因子对BALB/c小鼠T淋巴细胞免疫功能的促进作用及其作用机理。方法(1)体外,以正常T淋巴细胞、环磷酰胺所致免疫抑制T淋巴细胞、~(60)Coγ射线辐射所致免疫抑制T淋巴细胞为GPIF药效学研究模型,MTT法测定ConA诱导的T淋巴细胞的增殖反应;直接免疫荧光抗体标记,FACS分析脾T淋巴细胞、胸腺淋巴细胞表达CD3、CD4、CD8、CD28、CD152阳性细胞百分率;ELISA法测定T淋巴细胞培养上清IL2、IFN-γ水平,间接法测定IL4的活性;(2)体内,分别建立环磷酰胺、~(60)Coγ射线辐射所致免疫抑制动物模型,以耳片差重法测定GPIF对小鼠DTH反应能力的影响;建立60Coγ射线辐射所致免疫抑制动物模型,直接免疫荧光抗体标记、FACS分析GPIF对脾淋巴细胞、胸腺细胞表达CD3、CD4、CD8的影响,FACS分析GPIF对小鼠脾T淋巴细胞及其CD4~+、CD8~+亚群T淋巴细胞表达CD28、CD152的影响;ELISA法测定小鼠血清IL2、IFN-γ生成水平,间接
[Abstract]:According to the different effects of drugs on immune function, immunopharmacologists generally divide immune modulators into two categories: immune enhancers and immunosuppressants. The effect of the former is to promote the immune function, while the latter is the inhibition of the immune response. Goat placenta immunomodulatory factor is a bioactive substance prepared from goat placenta by modern biological technology in our laboratory. The preliminary activity analysis shows that it has immune enhancement activity. On the basis of analyzing and summarizing the current research data on the characteristics of immunological enhancement of animal derived immunoreactive peptides at home and abroad, this study was based on the principles of pharmacodynamics of immunoenhancers. The immune promoting effect and functional characteristics of goat placental immunomodulatory factor were systematically discussed, and the mechanism of its action was analyzed and studied.
