口蹄疫重组蛋白疫苗的计算机辅助设计及功能研究

【摘要】: 口蹄疫( Foot and mouth disease , FMD )是由口蹄疫病毒(Foot-and-mouth disease virus,FMDV)引起的一种急性高度传染性的疾病。由于其具有高度传染性和对农业经济的重大影响,国际兽医局(OIE)将口蹄疫列为A类传染病。 虽然接种口蹄疫灭活疫苗是预防和控制FMD爆发流行的有效手段,但因其在生产时易造成病毒泄漏,病毒灭活不全,有病毒扩散的隐患,促使我们寻求一种安全有效的疫苗来替代它,重组蛋白疫苗无疑是很好选择。 随着计算机技术的发展,表位预测、蛋白质结构预测使疫苗的设计有据可循。我们运用计算机技术设计了AsiaⅠ型口蹄疫疫苗RA2,并用基因工程的手段构建了重组牛AsiaⅠ型口蹄疫疫苗RA2的编码基因,在大肠杆菌中表达,并检验了该重组疫苗的免疫效果。 免疫豚鼠的实验表明,RA2疫苗在豚鼠体内可诱生高水平的抗AsiaⅠFMDV的中和性抗体。攻毒实验表明,RA2疫苗免疫的牛有3/5得到了保护,提示RA2可能成为抗牛AsiaⅠ型口蹄疫病毒的有效疫苗。 综上所述,采用计算机辅助设计,可以以较少的工作量构建出免疫效果较好的疫苗。
[Abstract]:Foot and mouth disease (Foot and mouth disease, FMD) is an acute infectious disease caused by foot and mouth disease virus (Foot-and-mouth disease virus,FMDV). Because of its high infectivity and great impact on agricultural economy, (OIE) of International Veterinary Bureau classified foot-and-mouth disease as Class A infectious disease. Although vaccination with inactivated vaccine against foot-and-mouth disease is an effective means to prevent and control the outbreak of FMD, it is easy to cause virus leakage and inactivation of the virus during production. In order to find a safe and effective vaccine to replace it, recombinant protein vaccine is undoubtedly a good choice.
