旋毛虫抗A549肿瘤效应的研究

【摘要】: 通过对BalB/C小鼠接种以不同方式处理的旋毛虫及不同旋毛虫抗原,再于不同时间荷以A549肺癌细胞,经对试验小鼠体内肿瘤的形态学指标检测、小鼠死亡率及其与抑瘤率的分析,观察旋毛虫对A549肺癌细胞的抑制作用。此外,在ELISA初步确定A549肺癌细胞抗原与旋毛虫抗原是否存在免疫交叉反应基础上,通过SDS-PAGE、Western-blot进一步确定两者相关抗原分子量大小,最后利用免疫组化对相关抗原进行定位。 研究结果发现未处理的旋毛虫抑瘤效果最佳(P=0.005),接种11d后荷瘤组的抑瘤效果好于接种7d前荷瘤组(P=0.006);旋毛虫ES抗原的抑瘤效果好于旋毛虫可溶性粗抗原(P=0.001);旋毛虫与A549肺癌细胞间相关抗原分子量大小为97.4、68KDa;经免疫组化定位分析确定相关抗原产生的抗体可结合于A549肺癌细胞的表面。
[Abstract]:BalB/C mice were inoculated with Trichinella spiralis and different antigens of Trichinella spiralis in different ways and then charged with A549 lung cancer cells at different time. To observe the inhibitory effect of Trichinella spiralis on A549 lung cancer cells. In addition, the molecular weight of A549 lung cancer cell antigen and Trichinella spiralis antigen were further determined by SDS-PAGE,Western-blot on the basis of the preliminary determination of A549 lung cancer cell antigen and trichinella antigen by ELISA. The results showed that the antitumor effect of untreated Trichinella spiralis was the best (P0. 005). After 11 d inoculation, the inhibition effect of Trichinella spiralis was better than that of P0. 006 before inoculation, the inhibitory effect of Trichinella spiralis ES antigen was better than that of soluble crude antigen of Trichinella spiralis (P0. 001), and that of Trichinella spiralis and A549 was better than that of P0. 006 before inoculation. The molecular weight of the intercellular associated antigen of lung cancer was 97.446 ~ 68KDa.The antibody produced by the relevant antigen could bind to the surface of A549 lung cancer cell by immunohistochemical localization analysis.
