Ghrelin对大鼠消化间期胃肠肌电活动的影响及其受体在消化道的分布特征

【摘要】:目的探讨ghrelin对大鼠消化间期复合肌电活动(IMC)的影响及其机制。方法采用免疫组织化学和图像分析方法观察ghrelin受体(生长激素促分泌素受体,GHS-R)在大鼠消化道的分布特征;采用多导生理记录仪监测大鼠消化间期IMC,观察静脉给予ghrelin对胃肠IMC的影响;采用放射免疫法测定静脉给予ghrelin前后,IMC不同时相的血浆胃动素浓度。结果①GHS-R在胃肠道的肠肌丛和黏膜下丛有表达,胃腺体均有GHS-R免疫强阳性广泛表达,在肠腺体的阳性表达较少。②静脉给予ghrelin促进胃肠IMC,IMC周期缩短,Ⅲ相时程缩短,Ⅲ相占IMC周期百分比无显著性改变;静脉注射ghrelin后,血浆胃动素周期性波动规律不受影响,各相应时相胃动素浓度与ghrelin作用前比较无显著性差异。结论 GHS-R主要分布于大鼠消化道的肠肌丛和黏膜下丛,在胃腺体有免疫强阳性表达,在小肠腺有少量阳性表达。Ghrelin可促进胃肠运动,此作用与胃动素没有明显关系。
[Abstract]:Objective to investigate the effect and mechanism of ghrelin on interdigestive complex myoelectric activity (IMC) in rats. Methods Immunohistochemistry and image analysis were used to observe the distribution of ghrelin receptor (GHS-R) in the digestive tract of rats. The effects of intravenous administration of ghrelin on gastrointestinal IMC in rats were observed by multichannel physiological recorder during interdigestive period, and plasma motilin concentrations at different phases before and after ghrelin administration were measured by radioimmunoassay. Results 1GHS-R was expressed in myenteric plexus and submucosal plexus of gastrointestinal tract, and GHS-R immunoreactivity was widely expressed in gastric glands. The positive expression of ghrelin in intestinal gland was less than that in intestinal gland.
