猫备用背根节内NT-3和相关神经肽的表达及其作用探讨

【摘要】:在胚胎早期神经营养素-3(neurotrophin-3,NT-3)对神经元的存活、突起生长及靶选择发挥着重要的作用,随着胚胎的发育其表达逐渐减少,功能作用也随之减弱。在成年动物神经损伤后,NT-3的表达再次增高,,参与到损伤修复的过程中。有实验显示成年猫部分背根损伤后备用背根节中NT-3mRNA表达上调,同时保留的备用背根在脊髓内发生了侧支出芽,以代偿丢失的初级传入纤维,提示增多的NT-3很可能参与了上述的代偿、修复过程。 为了探明成年猫正常背根节和部分去背根后备用背根节内源性NT-3对节内神经元的作用和作用模式,NT-3对备用背根侧支出芽的影响。以及背根节内与NT-3功能可能有相关关系的神经肽的表达及作用,本文采用CTB神经束路追踪、分离神经元培养、免疫组织化学、免疫荧光、酶联免疫吸附等技术从三个方面进行研究:NT-3促背根节神经突起生长作用探讨;备用背根节内NT-3表达的变化及作用模式探讨:背根节神经元内NT-3与肽类物质的共存及相关作用的探讨。
[Abstract]:Neurotrophin-3 (neurotrophin-3,NT-3) plays an important role in neuronal survival, process growth and target selection in early embryonic development. After nerve injury in adult animals, the expression of NT-3 was increased again and involved in the process of injury and repair. Some experiments showed that the expression of NT-3mRNA was up-regulated in the spare dorsal root ganglion after partial dorsal root injury in adult cats, while the reserved dorsal root developed lateral sprouts in the spinal cord to compensate for the lost primary afferent fibers. It is suggested that increased NT-3 may be involved in the above compensatory and repair process. In order to investigate the effect and mode of endogenous NT-3 on neurons in normal dorsal root ganglion (DRG) and partial dorsal root ganglia (DRG) of adult cats, the effect of NT-3 on spare dorsal root outgrowth bud was studied. The expression and role of neuropeptides in dorsal root ganglion which may be related to NT-3 function were studied by CTB nerve tract tracing, neuronal culture, immunohistochemistry and immunofluorescence. Enzyme-linked immunosorbent assay (Elisa) was used to study the effects of NT-3 on the growth of dorsal root ganglion (DRG) neurites. The changes of NT-3 expression in dorsal root ganglion and its mechanism of action: the coexistence of NT-3 and peptide in dorsal root ganglion neurons and the correlation between them.
