B淋巴细胞刺激因子及其受体与疾病关联性的研究

【摘要】:为探讨由B淋巴细胞刺激因子(BLyS)及其受体所介导的B细胞激活对疾病的致病机理,本研究根据Taqman技术的基本原理,在国内率先建立了实时定量检测BLyS受体mRNA含量的方法,并在国内外首先对系统性红斑狼疮(SLE)和多发性骨髓瘤(MM)患者血清BLys蛋白和外周血单个核细胞(peripheral blood mononuclear cells,,PBMCs)BLys及其受体mRNA表达水平进行检测,初步探讨BLys及其受体在SLE和MM患者发病的作用;同时将检测结果与两个疾病其他临床指标进行相关性研究,以探讨BLyS及其受体在相关疾病的预后及治疗等方面的意义;因为BAFF-R为BLyS主要和特异性受体,我们构建人BAFF-R 5′侧翼区5′系列缺失的重组质粒,为进一步研究BAFF-R基因转录调控机制奠定基础。
[Abstract]:In order to investigate the pathogenetic mechanism of B cell activation mediated by B lymphocyte stimulating factor (BLyS) and its receptor, a method of real-time and quantitative detection of BLyS receptor mRNA was established in China according to the basic principle of Taqman technique. The expression levels of serum BLys protein, peripheral blood mononuclear cell (peripheral blood mononuclear cells,PBMCs) BLys and its receptor mRNA in patients with systemic lupus erythematosus (SLE) and multiple myeloma (MM) were detected at home and abroad. To explore the role of BLys and its receptors in the pathogenesis of SLE and MM, and to study the correlation between the results of BLyS and other clinical indicators of the two diseases, and to explore the significance of BLyS and its receptors in the prognosis and treatment of related diseases. Because BAFF-R is the main and specific receptor for BLyS, we constructed the recombinant plasmid of 5 '-deletion of human BAFF-R 5'flanking region, which laid a foundation for further study on the transcriptional regulation mechanism of BAFF-R gene.
