小鼠妊娠高血压综合征模型的建立及妊高征对子代发育影响的观察

【摘要】:妊娠高血压综合征(妊高征)是一种常见的妊娠并发症,大约影响到5-10%的孕妇,仍然是妇产科导致孕产妇和胎儿/新生儿发病和死亡的主要原因。尽管进行了广泛的研究,引起妊高征的病因学因素还不完全清楚。建立合适的动物模型模仿人类妊高征,有助于人们了解妊高征的病理发生和发展进程,帮助理解与妊高征有关的病因学因素,建立临床上的诊断指标,筛选治疗药物具有重大意义;还可以详细研究妊高征对子代发育的影响,为提前介入治疗提供实验依据。 近年来环境因素在疾病发生过程中的重要作用受到了医学工作者和科学家的广泛注意。为更好地理解环境因素对妊高征的影响,研究者们试图把动物置于不同的环境条件下诱导产生实验性高血压,了解这些因素对妊娠的影响。高盐饮食与高血压存在相关性已经得到广泛认可,而冬季是妊高征高发季节的事实也说明寒冷刺激可能是致病因素之一。我们拟选择NaCl饲喂和寒冷刺激分别作为建模手段,以广泛使用的昆明种小鼠为材料,建立小鼠妊高征模型并观察妊高征对子代发育的影响。 饲喂诱导,分别使用1%、1.5%、2%三种浓度的NaCl溶液和饮水对照设四组小鼠,,连续饲喂14周。于14周术颈动脉插管法检测各组血压,确定饲喂浓度。以产生高血压组的NaCl浓度重新实验,连续饲喂14周后,安排雌雄合笼检出交配小鼠,设立正常对照、正常妊娠、饲喂对照、饲喂妊娠四组,在妊娠18天时测量各组小鼠血压,解剖孕鼠检查妊娠状况。 寒冷刺激诱导,将小鼠放入4±2℃冷室,分别每天刺激2h、4h、6h,在第18天时检测各组血压,与正常对照组比较找出每天的最佳刺激时间。以每日最佳刺激时间分别于冷激6、12、18天检测刺激期间小鼠的血压变化情况,了解冷激效应产生的时间。随后,设立正常对照、冷激对照、正常妊娠、冷激妊娠四组进行妊娠试验,在孕鼠妊娠18天时,各组小鼠称体重、麻醉,测量血压,取血保存血浆以备检测ATⅡ;解剖孕鼠,固定母鼠肾脏做切片检查,
[Abstract]:Pregnancy-induced hypertension syndrome (PIH) is a common complication of pregnancy affecting about 5-10% of pregnant women. It is still the main cause of maternal and fetal / neonatal morbidity and mortality in obstetrics and gynaecology. Despite extensive research, the etiology of PIH is not fully understood. Establishing suitable animal models to imitate human PIH can help people to understand the pathological process of PIH, to understand the etiological factors related to PIH, and to establish clinical diagnostic indexes. Screening treatment drugs is of great significance, and the influence of PIH on the development of offspring can be studied in detail to provide experimental basis for early interventional therapy. In recent years, the important role of environmental factors in the process of disease has been widely paid attention by medical workers and scientists. To better understand the effects of environmental factors on PIH, the researchers tried to induce experimental hypertension in animals under different environmental conditions to understand the effects of these factors on pregnancy. The association between high salt diet and hypertension has been widely accepted, and the fact that winter is the season of PIH also suggests that cold stimulation may be one of the pathogenic factors. We choose NaCl and cold stimulation as modeling methods to establish the model of PIH and observe the effect of PIH on the development of offspring in Kunming mice. Four groups of mice were fed with 1: 1. 5% and 2% NaCl solution and drinking water respectively. The mice were fed continuously for 14 weeks. Blood pressure was measured by carotid artery catheterization at 14 weeks and feeding concentration was determined. The concentration of NaCl in hypertension group was retested. After 14 weeks of continuous feeding, mating mice were detected in female and male cage, normal control group, normal pregnancy group, control group and pregnancy group were fed.
