STR基因座群体遗传学的研究及数据分析软件的开发与应用

【摘要】:目的 研究D15S659和D9S925基因座的群体遗传学和等位基因序列特征,积累群体遗传学数据,,探讨其法医学应用价值,为法医遗传学提供新的候选遗传标记。 方法 以109名华东地区汉族无血缘关系个体的静脉血为研究样本。用Chelex法提取DNA,对D15S659和D9S925基因座进行PCR扩增;非变性聚丙烯酰胺凝胶连续缓冲系统垂直电泳进行分型。利用Modified-Powerstats标准版软件对分型结果进行统计分析,获得各基因座的等位基因频率、基因型计数及法医群体遗传学数据。 结果 D15S659和D9S925基因座在华东汉族群体中等位基因数分别为8和6;杂合度分别为0.826和0.798;个人识别率分别为0.944和0.894;非父排除率分别为0.648和0.596。 结论 在我国华东地区汉族人群中,D15S659和D9S925基因座等位基因频率分布较好,提供的多态信息量大。具有较高的非父排除率和个人识别能力,适合于法医学应用,是群体遗传学研究和法科学应用较理想的候选STR基因座。
[Abstract]:Objective to study the population genetics and allelic sequence characteristics of D15S659 and D9S925 loci, accumulate population genetic data, and explore the value of forensic application to provide new candidate genetic markers for forensic genetics. Methods the venous blood samples were collected from 109 unrelated individuals of Han nationality in East China. The D15S659 and D9S925 loci were amplified by PCR with DNA, extracted by Chelex method, and typed by vertical electrophoresis of nondenatured polyacrylamide gel continuous buffer system. The allele frequency, genotype count and forensic population genetic data of each locus were obtained by statistical analysis of typing results with Modified-Powerstats standard software. Results the alleles of D15S659 and D9S925 loci were 8 and 6, heterozygosity were 0.826 and 0.798, personal identification rates were 0.944 and 0.894, non-paternal exclusion rates were 0.648 and 0.596, respectively. Conclusion the allelic frequencies of D15S659 and D9S925 loci are well distributed and the amount of polymorphic information is large in the Han population in East China. It has high non-paternal exclusion rate and personal recognition ability and is suitable for forensic application. It is an ideal candidate STR locus for population genetics and forensic science.
