CD40信号激活对肾小管上皮细胞的影响

【摘要】:目的探讨CD40信号激活对肾小管上皮细胞(HK2)的作用。方法以不同剂量的CD40激发型单克隆抗体抗(5c11)刺激HK2细胞,以四甲基偶氮唑盐(MTT)法和流式细胞术检测细胞增殖和凋亡,以酶联免疫吸附法检测HK2培养中趋化因子受激活调节正常T细胞表达和分泌因子(RANTES)水平。结果 HK2表面CD40表达高达78.9%。在不同浓度5c11刺激下,第9天时随5c11浓度增加HK2细胞增殖(OD值)逐步下降,各组间的差异均有统计学意义(P0.01);第6、第9天时HK2凋亡率升高,差异有统计学意义(P0.01),RANTES分泌随5c11浓度增加而升高,差异有统计学意义(P均0.05),并随时间推移呈增加趋势,差异有统计学意义(P均0.01)。结论 CD40信号在HK2细胞中的表达在导致肾固有细胞凋亡以及效应性细胞因子的分泌过程中起重要调节作用。
[Abstract]:Objective to investigate the effect of CD40 signal activation on renal tubular epithelial cells (HK2). Methods HK2 cells were stimulated with different doses of CD40 activated monoclonal antibody (5c11). The proliferation and apoptosis of HK2 cells were detected by (MTT) and flow cytometry. Enzyme linked immunosorbent assay (Elisa) was used to detect the level of normal T cell expression and secretion factor (RANTES) in HK2 culture. Results the expression of CD40 on HK2 was 78.9%. On the 9th day, the proliferation (OD) of HK2 cells decreased gradually with the increase of 5c11 concentration under different concentrations of 5c11, and the differences among each group were statistically significant (P0.01). On the 6th and 9th day, the apoptosis rate of HK2 increased, and the difference was statistically significant (P0.01), RANTES secretion increased with the increase of 5c11 concentration (P0. 05). The difference was statistically significant (P 0.01). Conclusion the expression of CD40 signal in HK2 cells plays an important role in the apoptosis of renal intrinsic cells and the secretion of effector cytokines.
