高糖诱导的星形胶质细胞凋亡的氧化应激通路及总三帖酸的干预作用

【摘要】:目的研究总三帖酸对高糖诱导的星形胶质细胞凋亡的干预作用及机制。方法采用体外分离乳鼠星形胶质细胞培养并分为对照组、模型组、总三帖酸干预组。高糖温孵后提取细胞mRNA和蛋白质分析细胞凋亡蛋白Bax/Bcl2及NADPH氧化酶亚基p22的表达。结果模型组细胞Bax表达上调而Bcl2表达下调,同时p22表达上调。总三萜酸干预后上述蛋白表达部分得到恢复。结论总三帖酸可通过抑制p22上调抑制高糖引起的星形胶质细胞凋亡,减轻细胞的损伤。
[Abstract]:Objective to study the effect and mechanism of total tricitric acid on apoptosis of astrocytes induced by high glucose. Methods astrocytes were isolated from neonatal rats in vitro and divided into three groups: control group, model group and total tricitric acid intervention group. MRNA and protein expression of apoptotic protein Bax/Bcl2 and NADPH oxidase subunit p22 were analyzed. Results in the model group, the expression of Bax was up-regulated, the expression of Bcl2 was down-regulated, and the expression of p22 was up-regulated. After total triterpene acid intervention, the above protein expression was partially recovered. Conclusion TSA can inhibit the apoptosis of astrocytes induced by high glucose by inhibiting the up-regulation of p22 and attenuating the injury of astrocytes.
