大鼠机械性脑损伤蛋白质表达谱的变化及意义

【摘要】:目的:研究闭合性颅脑损伤大鼠血清和海马蛋白质表达谱,探寻脑损伤生物标志蛋白。 方法:选雄性SD 大鼠随机分为手术对照组及损伤后4 h、8 h、12 h、24 h 和48 h 组,复制Marmarou’s 落体打击大鼠脑损伤模型。分别进行神经行为学评价、常规HE 染色、尼氏染色和酯化银染色;并用弱阳离子交换芯片(WCX2)和金属亲和吸附芯片(IMAC3)结合表面增强激光解析电离飞行时间质谱技术分析大鼠闭合性脑损伤后不同时间血清和海马中蛋白质表达谱的改变。 结果:1. 临床表现:损伤组大鼠于打击后有84 只大鼠(70%)出现抽搐同时有呼吸暂停,持续约15s;在没有出现抽搐的大鼠中有11 只大鼠(9.17%)出现惊厥,持续约30s;17 只大鼠(14.3%)伤后有肢体活动受限,表现为单肢或单侧肢体活动受限。12 只大鼠(10%)于打击后出现持续呼吸抑制和全身抽搐,几十秒钟后呼吸停止,死亡。 2. 神经行为学评价:大鼠脑损伤后平衡能力、翻身、支头、行走、逃避和四肢功能均较损伤前有所减弱,评分降低。脑损伤后1 h、12 h 大鼠行为学评分与损伤前相比有显著性差异(P0.01),损伤后24 h、48 h 与损伤前比较无显著性差异。 3. 大体病理:损伤组大鼠可见蛛网膜下腔出血,分布广泛,无明显大面积挫伤或局灶性挫裂伤;切面可见胼胝体点状出
[Abstract]:Aim: to study the protein expression profiles of serum and hippocampus in rats with closed brain injury and to explore the biomarkers of brain injury. Methods: male SD rats were randomly divided into two groups: the control group and the control group. The rats were randomly divided into two groups: the control group and the 24 h and 48 h groups after 4 h, 8 h and 12 h after injury. The model of brain injury was induced by Marmarou's falling body. Neurobehavioral evaluation, routine HE staining, Nissl staining and esterification silver staining were performed. The changes of protein expression profiles in rat serum and hippocampus at different times after closed brain injury were analyzed by weakly cation exchange chip (WCX2) and metal affinity adsorption chip (IMAC3) combined with surface enhanced laser desorption ionization time of flight mass spectrometry. The result is 1: 1. Clinical manifestations: 84 rats (70%) in the injury group had convulsions and apnea for about 15 s, 11 rats (9.17%) showed convulsion for about 30 s. 17 rats (14.3%) had limited limb movement after injury, which was characterized by limited movement of one or one limb, 12 rats (10%) had sustained respiratory inhibition and systemic convulsions after striking, and after dozens of seconds, breathing stopped and died. 2. Neurobehavioral evaluation: after brain injury, the ability of balance, turning over, branching, walking, escaping and limb function were all weakened and the score was lower. There was a significant difference in behavioral score between 1 hour and 12 h after brain injury (P0.01), but there was no significant difference between 24 h and 48 h after injury. 3. Gross pathology: subarachnoid hemorrhage was found in the injury group, with no obvious large area contusion or focal contusion, and corpus callosum punctate on the section.
