大鼠机械性脑损伤生物标志物的研究

【摘要】: 目的:研究闭合性颅脑损伤大鼠血清和海马蛋白质表达谱,探寻脑损伤生物标志蛋白。 方法:选雄性SD大鼠随机分为手术对照组及损伤后4 h、8 h、12 h、24 h和48 h组,复制Marmarou’s落体打击大鼠脑损伤模型。分别进行常规HE染色、尼氏染色;并用强阴离子交换芯片(SAX2)和疏水表面芯片(H4)结合表面增强激光解析电离飞行时间质谱技术分析大鼠闭合性脑损伤后不同时间血清和海马中蛋白质表达谱的改变。 结果:1临床表现损伤组大鼠于打击后有84只大鼠(70%)出现抽搐同时有呼吸暂停,持续约15s;在没有出现抽搐的大鼠中有11只大鼠(9.17%)出现惊厥,持续约30s;17只大鼠(14.3%)伤后有肢体活动受限,表现为单肢或单侧肢体活动受限。12只大鼠(10%)于打击后出现持续呼吸抑制和全身抽搐,几十秒钟后呼吸停止,死亡。 2大体病理损伤组大鼠可见蛛网膜下腔出血,分布广泛,无明显大面积挫伤或局灶性挫裂伤;切面可见胼胝体点状出血及双侧脑室出血。 3光镜下观察损伤组大鼠HE染色主要表现为额叶、顶叶、枕叶、小脑及脑干蛛网膜下腔出血,侧脑室出血,小挫伤灶。部分神经细胞胞体浓缩,胞核固缩,胞浆内尼氏小体消失呈同质性嗜酸性着色(红色神经细胞);部分神经细胞胞体肿胀,胞浆不规则淡染,核肿大淡染或消失。脑组织水肿,表现
[Abstract]:Aim: to study the protein expression profiles of serum and hippocampus in rats with closed brain injury and to explore the biomarkers of brain injury. Methods: male SD rats were randomly divided into two groups: the control group and the control group. The rats were randomly divided into two groups: the control group and the 24 h and 48 h groups after 4 h, 8 h and 12 h after injury. The model of brain injury was induced by Marmarou's falling body. Routine HE staining and Nissl staining were performed respectively. Strong anion exchange chip (SAX2) and hydrophobic surface chip (H4) combined with surface enhanced laser desorption ionization time of flight mass spectrometry were used to analyze the changes of protein expression profiles in rat serum and hippocampus at different time after closed brain injury. Results: 1 there were 84 (70%) rats (70%) with convulsions and apnea (15 s) in the clinical manifestation group, 11 (9.17%) of the rats (9.17%) had convulsions and 30 s of convulsions. 17 rats (14.3%) had limited limb movement after injury, which was characterized by limited movement of one or one limb, 12 rats (10%) had sustained respiratory inhibition and systemic convulsions after striking, and after dozens of seconds, breathing stopped and died. 2Subarachnoid hemorrhage was found in the gross pathological injury group, with no obvious large area contusion or focal contusion, and corpus callosum punctate hemorrhage and bilateral ventricular hemorrhage were observed on the cut surface. 3The HE staining of the injured rats under light microscope was mainly manifested as subarachnoid hemorrhage in frontal lobe, parietal lobe, occipital lobe, cerebellum and brainstem, lateral ventricular hemorrhage and small contusion. Some of the neuronal bodies were concentrated, the nuclei were constricted, and the Nissl bodies disappeared in the cytoplasm with homogeneous eosinophilic staining (red nerve cells), some of the neuronal cells were swollen, irregularly stained, and the nuclei were dilated or disappeared.
