以EGFR为靶点siRNA的初步研究

【摘要】: 表皮生长因子受体(epidermal growth factor receptor, EGFR)的过度表达被认为是肿瘤细胞过度增殖的重要因素之一。抑制EGFR的过度表达可以有效抑制细胞的过度增殖,从而达到肿瘤临床治疗的目的。RNA干扰(RNA interference, RNAi)技术可以高效高特异性地抑制特异蛋白的表达量,在肿瘤的治疗方面具有良好的应用前景。 本研究在体外培养角质形成细胞和舌鳞癌细胞的基础上,根据小干扰RNA(small interfering RNA, siRNA)的设计原则设计并合成以EGFR为靶点的siRNA,通过RNAi技术研究其对角质形成细胞和舌鳞癌细胞EGFR mRNA表达的抑制作用。从实验结果中筛选出一条高效的以EGFR为靶点siRNA。研究其对舌鳞癌细胞增殖的抑制作用。该研究为以EGFR为靶点的siRNA临床治疗鳞癌、银屑病等EGFR过度表达的临床疾病研究提供一种新的实验方法,为以RGFR为靶点药物在临床疾病治疗中的应用提供一定的实验依据。
[Abstract]:The overexpression of epidermal growth factor receptor (epidermal growth factor receptor, EGFR) is considered to be one of the important factors in the proliferation of tumor cells. Inhibiting the overexpression of EGFR can effectively inhibit the excessive proliferation of cells and achieve the purpose of clinical treatment of tumor. RNA interference (RNA interference, RNAi) technique can inhibit the expression of specific protein efficiently and specifically. It has a good prospect in the treatment of tumor. In this study, keratinocytes and tongue squamous carcinoma cells were cultured in vitro, and siRNA, targeting EGFR was designed and synthesized according to the design principle of small interference RNA (small interfering RNA, siRNA). The inhibition of EGFR mRNA expression in keratinocytes and tongue squamous carcinoma cells was studied by RNAi technique. A highly efficient siRNA. targeting EGFR was selected from the experimental results. To study its inhibitory effect on the proliferation of tongue squamous cell carcinoma cells. This study provides a new experimental method for the clinical treatment of squamous cell carcinoma, psoriasis and other overexpressed EGFR diseases with siRNA targeted at EGFR, and provides a certain experimental basis for the application of RGFR as a target drug in the treatment of clinical diseases.
