内毒素血症大鼠脑底动脉内皮素-1表达的变化

【摘要】:目的:探讨内毒素血症大鼠血浆ET-1水平、脑底动脉内皮素-1(Endothelin-1,ET-1)能神经纤维诱发脑血管痉挛(Cerebral vasopasm,CVS)发生与发展的关系,为深入阐明内毒素血症诱发CVS的发病机制提供理论依据。方法:96只Wistar大鼠随机分为正常对照组、内毒素血症组(3、6、12、24、48h),分别用放射免疫分析(Radioimmunoassay,RIA)技术、免疫组织化学ABC法(Immunohistochemical technique ABC)对正常对照组、内毒素血症组(3、6、12、24、48h)血浆ET-1水平表达的变化及大鼠脑底动脉ET-1能神经纤维进行观察。结果:内毒素血症后3h、6h、12h大鼠血浆ET-1水平比正常对照组明显升高(P<0.05),内毒素血症后24h和48h趋于正常(P>0.05);正常对照组大鼠大脑前动脉、大脑中动脉、大脑后动脉和基底动脉均可见棕褐色,呈细线状ET-1能免疫反应阳性纤维;内毒素血症后6h、12h组大鼠脑底动脉与正常对照组大鼠脑底动脉比较,ET-1能阳性神经纤维密度明显增多,尤其以内毒素血症后12h组大鼠脑底动脉ET-1能神经纤维密度改变最为明显,经统计学检验(P<0.05),内毒素血症后48h组大鼠脑底动脉ET-1能神经纤维密度无明显变化(P>0.05)。结论:内毒素血症大鼠血浆ET-1水平升高、脑底动脉ET-1能神经纤维密度增加,提示ET-1参与了内毒素血症诱发脑血管痉挛的病理生理过程,表明ET-1在内毒素血症诱发脑血管痉挛中起重要作用,,揭示了脑血管痉挛的神经-体液调节机制的新思路,同时为临床预防和治疗脑血
[Abstract]:Objective: to investigate the level of plasma ET-1 in rats with endotoxemia and the relationship between the occurrence and development of cerebral vasospasm (Cerebral vasopasm,CVS) induced by endothelin-1 (Endothelin-1,ET-1) nerve fibers in the basal cerebral artery. To provide a theoretical basis for elucidating the pathogenesis of CVS induced by endotoxemia. Methods: 96 Wistar rats were randomly divided into two groups: normal control group, endotoxemia group (36 / 24 h). Radioimmunoassay (Radioimmunoassay,RIA) technique and immunohistochemical ABC method (Immunohistochemical technique ABC) were used to treat normal control group and endotoxemia group (3 / 6), respectively. The changes of plasma ET-1 expression and ET-1 nerve fibers in rat cerebral artery were observed. Results: the level of plasma ET-1 in rats was significantly higher than that in the control group at 3h, 6h and 12h after endotoxemia (P < 0. 05), and tended to be normal at 24h and 48h after endotoxemia (P > 0. 05). In the control group, the anterior cerebral artery, middle cerebral artery, posterior cerebral artery and basilar artery were all brown, with thin line ET-1 immunoreactive fibers. Compared with the normal control group, the density of ET-1 positive nerve fibers increased significantly in the cerebral fundus artery of rats 6 h after endotoxemia and 12 h after endotoxemia. The density of ET-1 nerve fibers in the cerebral fundus artery of rats in 12h after endotoxemia was the most obvious (P < 0. 05). There was no significant change in the density of ET-1 nerve fibers in the cerebral fundus artery of rats 48 hours after endotoxemia (P > 0. 05). Conclusion: the level of plasma ET-1 and the density of ET-1 energy nerve fibers in the basal cerebral artery are increased in rats with endotoxemia, suggesting that ET-1 is involved in the pathophysiological process of cerebral vasospasm induced by endotoxemia. The results suggest that ET-1 plays an important role in vasospasm induced by endotoxemia. It also reveals a new idea of neurohumoral regulation in cerebral vasospasm, and it also provides a new approach for clinical prevention and treatment of cerebral blood.
