青春双歧杆菌对Ⅰ型变态反应的影响

【摘要】: 目的观察青春双歧杆菌对Ⅰ型变态反应的抑制作用。 方法1、大鼠实验:1)以卵白蛋白为变应原致敏大鼠,制备主动致敏的腹腔肥大细胞。2)选用体外抗卵白蛋白主动致敏的大鼠腹腔肥大细胞为细胞模型,分为实验组和模型组。分别进行(1)甲苯胺蓝染色法观察青春双歧杆菌对体外主动致敏的肥大细胞脱颗粒的影响。(2)荧光分光光度法测定青春双歧杆菌对体外主动致敏的大鼠腹腔肥大细胞释放组胺的影响。(3)免疫组化法检测实验组和模型组中主动致敏的肥大细胞中5-HT的表达。2、小鼠实验:以卵白蛋白为变应原,建立Ⅰ型变态反应的实验动物模型,并将模型动物分为实验组和模型组,分别灌胃青春双歧杆菌活菌液和生理盐水,13天后,取静脉血,用流式细胞仪检测CD23的表达情况。 结果实验组和模型组中,肥大细胞脱颗粒的百分率分别是42.52±2.94%和90.89±1.47%,5-HT的染色积分分别是285.7±12.85和197.6±11.37,肥大细胞释放组胺的量分别是1.09±0.01ng/ml和1.30±0.06ng/ml,CD23的百分率分别是17.71±2.47%和27.44±2.49%,实验组和模型组相比有显著性差异(P<0.05)。光镜下观察:实验组细胞形态规则、脱颗粒细胞数减少。 结论双歧杆菌具有抑制肥大细胞脱颗粒的作用,抑制IgE的产生,从而减少Ⅰ型变态反应的发生。
[Abstract]:Objective to observe the inhibitory effect of Bifidobacterium pubertifolia on type I allergy. Methods 1, rat experiment: 1) using ovalbumin as allergen to sensitize rat peritoneal mast cells, and 2) using rat peritoneal mast cells which were anti-ovalbumin sensitized in vitro as cell model. It was divided into experimental group and model group. (1) toluidine blue staining method was used to observe the effect of bifidobacterium pubertifolia on degranulation of mast cells sensitized in vitro. (2) fluorescence spectrophotometry was used to determine the effect of bifidobacterium pubertifolia on the active sensitization of rat abdomen in vitro. (3) Immunohistochemical method was used to detect the expression of 5-HT in activated mast cells in experimental group and model group. Mouse experiment: using ovalbumin as allergen, the experimental animal model of type I allergic reaction was established, and the model animals were divided into experimental group and model group. Bifidobacterium puberty bifidobacterium solution and normal saline were perfused by stomach respectively. After 13 days, venous blood was taken. The expression of CD23 was detected by flow cytometry. Results the percentage of mast cell degranulation in experimental group and model group was 42.52 卤2.94% and 90.89 卤1.47 respectively. The staining scores of 5-HT were 285.7 卤12.85 and 197.6 卤11.37, respectively. The percentage of histamine released by mast cells was 1.09 卤0.01ng/ml and 1.30 卤0.06 ng / ml CD23, respectively, which were 17.71 卤2.47% and 27.44 卤2.49%, respectively. There was a significant difference between the experimental group and the model group (P < 0. 05). Under the light microscope, the cells in the experimental group were regular in shape and the number of degranulated cells was decreased. Conclusion Bifidobacterium can inhibit the degranulation of mast cells and the production of IgE, thus reducing the occurrence of type I allergic reaction.
