小鼠皮肤胚胎期NOS阳性神经末梢的分布及其与羊水接触关系的研究

【摘要】:【背景和目的】 皮肤广泛分布于脊椎动物和人体表面,其表皮的基底层内分布着大量的浅感受器和丰富的神经末梢。发育神经生物学研究表明,皮肤是脊神经感觉纤维的靶区,在胚胎发育期间背根神经节细胞的周围突伸向皮肤特定的层次内,形成密集的神经网络,说明皮肤的发育与周围神经感觉纤维的发育之间一定存在着某种密切的关系。然而有关胚胎发育时期皮肤细胞与发育中的神经末梢之间联系的建立,背根神经节细胞的周围突如何被诱导向靶区生长及调节因素等问题都不是很清楚。 一氧化氮(Nitric Oxide,NO)是一种在细胞内和细胞间具有高度活性的信使分子,广泛分布于生物体内各组织中,特别是神经组织中。关于NO在中枢和外周神经系统的分布特点已有许多研究报道,但对胚胎期小鼠周围神经末梢的分布和发育未见报道。 因此,我们利用逆行轴浆运输的原理将荧光金(fluorogold,FG)和辣根过氧化物酶(horse radish peroxidase;horseradish peroxidase,HRP)在体注射到胚胎的羊水中,观察胚胎体神经和背根神经节细胞是否含有荧光金或HRP,并通过
[Abstract]:[background and objective] skin is widely distributed on vertebrate and human surface, and a large number of superficial receptors and abundant nerve endings are distributed in the basal layer of the epidermis. Developmental neurobiology studies have shown that the skin is the target of the sensory fibers of the spinal nerve. During embryonic development, the processes around the dorsal root ganglion cells extend to a specific level of the skin, forming a dense neural network. It suggests that there must be a close relationship between the development of skin and the development of peripheral nerve sensory fibers. However, it is not clear that the relationship between the skin cells and the developing nerve endings during embryonic development, how the peripheral processes of dorsal root ganglion cells are induced to the target and the regulatory factors. Nitric oxide (Nitric Oxide,NO) is a highly active messenger molecule in cells and between cells, which is widely distributed in various tissues of organisms, especially in nerve tissues. The distribution of NO in central and peripheral nervous system has been reported, but the distribution and development of peripheral nerve endings in embryonic mice have not been reported. Therefore, we use the principle of retrograde axonal transport to transfer fluorogold,FG and horseradish peroxidase (horse radish peroxidase;.
