紫菜多糖对小鼠NK细胞杀伤活性和淋巴细胞转化功能的上调作用

【摘要】:前言 紫菜属于红藻类(Rhodophyta),红毛菜目(Bangiales),红毛菜科(Bangiaceae),紫菜属(Porphyra)。紫菜多糖(PP)是紫菜的重要组成成分,研究表明具有多种生物学功能,如抗凝血、降血脂、抗血栓、,抗炎、防治溃疡等作用,以及明显的抗突变、抗肿瘤作用。近年来紫菜多糖作为一种免疫调节剂逐渐被人们所认识,其可上调机体的免疫系统功能,刺激各种免疫活性细胞如自然杀伤细胞和B细胞成熟、分化和增殖,并能促进外周血淋巴细胞的转化,可以促进脾细胞分泌细胞毒效应分子TNF,使其细胞毒作用得到增强。 紫菜多糖主要由岩藻糖,半乳糖,甘露糖,葡萄糖,和木糖组成,是一种含糖醛酸的异多糖,典型的紫菜聚糖结构含有3,6-内醚-半乳糖基和硫酸基,研究表明,后者与紫菜多糖所具有的免疫调节作用关系密切。本实验通过给予小鼠腹腔注射紫菜多糖,取脾细胞检测NK细胞杀伤活性,淋巴细胞增殖功能,及产生TNF的含量,以探讨紫菜多糖对免疫功能的影响。 实验材料与方法 将干燥紫菜以蒸馏水浸湿,95%酒精脱脂,蒸馏水浸泡以除色素,以10倍水量文火煮沸提取,以三氯乙酸法(TCA法)除蛋白,无水乙醇沉淀多糖,冷冻干燥后得PP提取物。以苯酚-硫酸法检测纯度。将BALB/C小鼠随机分为4组,每组18只:生理盐水对照组,不同剂量PP处理组(0.05g·kg~(-1),0.25g·kg~(-1),0.5g·kg~(-1))连续免疫7天,分别于末次免疫后的24h,48h,72h,杀鼠各6只,无菌取脾,调细胞浓度至1×10~7/ml,用乳酸脱氢酶释放法检测NK细胞的活性,用MTT法检测淋巴细胞转化功能及脾培养上清中的TNF对靶细胞的杀伤活性,用SPSS统计软件对实验结果进行差异显
[Abstract]:Introduction Laver belongs to red algae (Rhodophyta), (Bangiales), (Bangiaceae), (Porphyra). Porphyra polysaccharide (PP) is an important component of porphyra. Studies have shown that it has a variety of biological functions, such as anticoagulant, antithrombotic, anti-inflammatory, anti-ulcer, as well as obvious anti-mutation, anti-tumor effects. In recent years, porphyra polysaccharide has been gradually recognized as an immunomodulator, which can up-regulate the function of the body's immune system and stimulate the maturation, differentiation and proliferation of various immune active cells such as natural killer cells and B cells. It can promote the transformation of peripheral blood lymphocytes and enhance the cytotoxicity of spleen cells by secreting cytotoxic response molecule TNF,. Porphyra polysaccharide, mainly composed of fucose, galactose, mannose, glucose, and xylose, is an isopolysaccharide containing uronic acid. The typical porphyra polysaccharide structure contains 3,6-internal ether-galactosyl and sulfate groups. The latter is closely related to the immunomodulatory effect of porphyra polysaccharide. In order to explore the effect of porphyra polysaccharide on immune function, the cytotoxicity of NK cells, the proliferation of lymphocytes and the production of TNF were detected by injecting porphyra polysaccharide into mice intraperitoneally. Materials and methods the dried porphyra was soaked in distilled water, degreased by 95% alcohol, soaked in distilled water to remove pigment, boiled in 10 times water and removed protein by trichloroacetic acid (TCA) method. The polysaccharide was precipitated by anhydrous ethanol and the PP extract was obtained after freeze-drying. The purity was determined by phenol-sulfuric acid method. BALB/C mice were randomly divided into 4 groups: saline control group (n = 18), PP treated group (0.05g kg~ (- 1), 0.25g kg~ (- 1), 0.5g kg~ (- 1) for 7 days. At 24 h, 48 h, 72 h after the last immunization, 6 mice in each group were killed. The spleen was taken aseptically and the cell concentration was adjusted to 1 脳 10 ~ 7 ml. The activity of NK cells was measured by lactate dehydrogenase release assay. The transformation function of lymphocytes and the killing activity of TNF in the supernatant of spleen culture against target cells were detected by MTT method. The difference of the experimental results was observed by SPSS statistical software.
