可分泌高活性β-半乳糖苷酶的重组乳酸乳球菌株的构建及其产酶特性的初步研究

【摘要】:目的 以高产β-半乳糖苷酶(β-galactosidase)的保加利亚乳杆菌(Lactobacillus bulgaricus)β-半乳糖苷酶基因(lacZ)为初始基因,递归PCR合成的乳酸乳球菌基因usp45为分泌信号肽基因,表达型质粒pMG36e为载体,构建重组质粒pMG36e-lacZ和pMG36e-usp-lacZ。电转化乳酸乳球菌MG1363,获得能分泌高活性β-半乳糖苷酶的重组乳酸乳球菌,并初步研究其产酶特性。为进一步构建食品级重组乳酸杆菌,用于乳糖酶缺乏的治疗和研究奠定基础。 方法 本研究分5个部分: 1.β-半乳糖苷酶基因初始菌的鉴定及其产酶特性研究:对基因初
[Abstract]:Objective to use the 尾-galactosidase (尾-galactosidase)-producing (Lactobacillus bulgaricus) 尾-galactosidase gene (lacZ) as the initial gene of Lactobacillus bulgaricus. The gene usp45 of Lactococcus lactis synthesized by recursive PCR was used as the secretory signal peptide gene, and the expression plasmid pMG36e was used as the vector. The recombinant plasmids pMG36e-lacZ and pMG36e-usp-lacZ. were constructed. The recombinant lactococcus, which can secrete high activity 尾-galactosidase, was obtained by electrotransformation of Lactococcus lactis MG1363,. It lays a foundation for the further construction of food-grade recombinant lactobacillus for the treatment and research of lactase deficiency. Methods this study was divided into five parts: 1.
