【摘要】:研究背景: 肺动脉高压是肺循环障碍和肺功能异常性疾病的常见表现,是肺心病发病机制的中心环节。急性和慢性低氧均可导致缺氧性肺动脉高压,其中肺血管收缩反应增强和肺血管结构改建是其主要血管变化特征。肺动脉平滑肌细胞(PASMC)的肥大增生是肺血管结构改建的主要表现之一,缺氧性PASMC增生是肺动脉高压形成的重要原因之一。 降钙素基因相关肽(Calcitonin gene-related peptide,CGRP)具有强大的舒血管作用,有研究报道缺氧性肺动脉高压大鼠的血浆CGRP水平明显下降,,且肺动脉压力与血浆CGRP浓度呈显著负相关,而给大鼠注射CGRP,则可减轻或逆转缺氧性肺动脉高压。 Rho GTP酶是真核细胞内重要的信号传递分子,在真核细胞信号传递过程中发挥着分子开关作用。Rho激酶是RhoA的下游效应蛋白,分子量约160kD,是一个Ser/Thr蛋白激酶。研究发现Rho激酶在凝血酶、血管紧张素Ⅱ、五羟色胺和Urotensin—Ⅱ等诱导的血管平滑肌细胞增生中
[Abstract]:Background: pulmonary hypertension is a common manifestation of pulmonary circulation disorder and abnormal pulmonary function, and is the central link of the pathogenesis of cor pulmonale. Both acute and chronic hypoxia can lead to hypoxic pulmonary hypertension, in which the enhancement of pulmonary vasoconstriction and the structural remodeling of pulmonary vessels are the main characteristics of pulmonary hypertension. Hypertrophic proliferation of pulmonary artery smooth muscle cells (PASMC) is one of the main manifestations of pulmonary vascular structural remodeling. Hypoxic PASMC proliferation is one of the important causes of pulmonary hypertension. Calcitonin gene-related peptide (Calcitonin gene-related peptide,CGRP) has a strong vasodilatory effect. It has been reported that the plasma CGRP level in hypoxic pulmonary hypertension rats is significantly decreased, and the pulmonary artery pressure is negatively correlated with the plasma CGRP concentration. Injection of CGRP, into rats could relieve or reverse hypoxic pulmonary hypertension. Rho GTP is an important signaling molecule in eukaryotic cells and plays a molecular switching role in eukaryotic signal transduction. Rho kinase is a downstream effector of RhoA with molecular weight of about 160kD.It is a Ser/Thr protein kinase.
