黄热疫苗病毒17D株在Vero细胞上的适应性传代

【摘要】:为建立黄热疫苗病毒17D株的Vero细胞适应株,将黄热疫苗病毒在Vero细胞上培养,通过蚀斑筛选,然后进行适应性传代,并进行稳定性研究。 将鸡胚黄热疫苗病毒在Vero细胞上进行了适应性培养,确定了传代培养条件,MOI为0.1 PFU/cell,接种方式为分种,培养温度为37℃。对2个批号的鸡胚黄热减毒活疫苗病毒进行了2次蚀斑筛选,并连续传代,获得了YFV-Vero(5)-A1、YFV-Vero(5)-A2、YFV-Vero(5)-B1和YFV-Vero(5)-B2等4株Vero细胞适应株病毒。经过传代后的稳定性研究表明,4株Vero细胞适应株病毒可被黄热病毒(17D)猴免疫血清中和,证实确为黄热17D株病毒。在传代过程中不同代次的病毒滴度稳定在6.9Lg PFU/ml以上,各代病毒蚀斑直径大小没有变化,高峰病变出现时间为5~7天。免疫原性研究结果表明,用BHK-21细胞和Vero细胞为基质进行中和试验,家兔和小鼠的免疫血清均可中和3批不同批号的鸡胚黄热减毒活疫苗,中和抗体效价为1:1280~1:5120,显示了4株Vero细胞适应株病毒良好的免疫原性。 初步试验结果表明,已获得的4株黄热减毒活疫苗Vero细胞适应株病毒,其病毒滴度高且稳定,中和抗体效价高,免疫原性良好。以后需进一步做猴体神经毒力试验以证实其遗传稳定性,做病毒株的全序列测定以分析其基因稳定性。
