CREST对N2a细胞突起生长的影响

【摘要】:目的观察钙反应性反式激活因子(CREST)过表达对小鼠神经母细胞瘤(N2a)细胞突起生长的影响。方法实验分为3组,实验组用pCMV-HA-CREST转染的N2a细胞,空载体对照组用pCMV-HA转染的N2a细胞,空白对照组用未转染任何试剂的N2a细胞,分别加入70mmol/L KCl孵育过夜。在转染后24h,48h采用考马斯亮蓝染色法检测N2a细胞突起生长状态。结果在转染后24h,CREST基因过表达的细胞与两对照组细胞相比较,细胞突起生长差异无显著性。在转染后48h,CREST基因过表达的细胞与两对照组细胞相比较,突起生长明显,部分突起交织成网络。两对照组间在细胞转染后各时间点突起生长状态差异无显著性(P0.05)。结论过表达CREST可促进N2a细胞突起的生长。
[Abstract]:Aim to investigate the effect of over-expression of calcium-responsive trans-activator (CREST) on neurite growth of mouse neuroblastoma (N2a) cells. Methods the experiment was divided into three groups: N2a cells transfected with pCMV-HA-CREST in experimental group, N2a cells transfected with pCMV-HA in empty vector control group, N2a cells transfected with pCMV-HA in blank control group and N2a cells transfected with no reagent in blank control group. 70mmol/L KCl was added to incubate N2a cells for the night. The neurite growth of N2a cells was detected by Coomassie brilliant blue staining at 24 h and 48 h after transfection. Results at 24 h after transfection, there was no significant difference in the neurite growth between the cells overexpressed by crest gene and those of the two control cells. At 48 h after transfection, the crest gene over-expressed cells grew significantly compared with the two control cells, and some of the processes were interwoven into a network. There was no significant difference in the neurite growth status between the two control groups at each time point after transfection (P0.05). Conclusion overexpression of CREST can promote the neurite growth of N2a cells.
