日本血吸虫云南品系和皖鄂品系对家兔致病性的比较研究

【摘要】:[目的] 观察和比较家兔在同等条件下分别感染日本血吸虫云南品系和皖鄂品系后,其肝功能的动态变化以及肝肠病理学改变和异位寄生情况,探讨不同地域的两种品系日本血吸虫对宿主致病性的差异。 [方法] 22只成年健康家兔随机分为对照组、大理组和皖鄂组,分别用大理钉螺和江苏钉螺逸出的血吸虫尾蚴(150±2条)以腹部贴片法攻击感染。用全自动生化分析仪于感染后不同时间分别检测对照组和实验组家兔血清总蛋白,白蛋白和球蛋白,丙氨酸氨基转移酶(ALT),天冬氨酸氨基转移酶(AST)及血清总胆红素和γ-谷氨酰转肽酶(γ-GT)等肝功能指标,并进行统计分析;于感染后不同时间分别剖杀实验动物,观察肝表面虫卵结节的密度及大小;在光学显微镜下观察肝脏、小肠和大肠等组织的病理学变化;用放射免疫法(RIA)检测感染后12W各组家兔血清透明质酸(HA),Ⅲ型前胶原(PCⅢ),Ⅳ型胶原(Ⅳ-C)等肝纤维化指标;压片观察肺脏和脑组织观察异位寄生情况。 [结果] 本实验中,实验组家兔血清白蛋白持续下降,而血清球蛋白持续上升,整个实验过程中血清总蛋白无明显变化,两种转氨酶及γ-谷氨酰转肽酶活性都以增高——进一步增高——恢复的形式变化,胆红素则一直升高。除胆红素外,其它的肝功能指标变化均以大理组为著。肝脏结构损害主要表现为:感染后2周,肝血窦扩张充血,肝细胞水肿,,少数肝细胞脂肪变性;感染后6周,门脉区有充血及炎细胞浸润,大部分成熟虫卵周围均有密集的以嗜酸性粒细胞为主的炎细胞浸润,有的则因炎细胞坏死崩解而形成急性虫卵脓肿;感染后12周,未见假小叶形成,汇管区有较多的虫卵结节,伴有纤维组织增生及慢性炎细胞浸润,小肠、结肠的病理改变为进行性加重的炎症反应,结肠可见不同时期的急性、
[Abstract]:[objective] to observe and compare the dynamic changes of liver function, pathological changes of liver and intestine and ectopic parasitism in rabbits infected with Schistosoma japonicum Yunnan strain and Anhui-Hubei strain under the same conditions, respectively. The pathogenicity of two strains of Schistosoma japonicum (Schistosoma japonicum) from different regions to host was studied. [methods] Twenty-two adult healthy rabbits were randomly divided into three groups: control group, Dali group and Anhui-Hubei group. The infected rabbits were attacked by abdominal patch method with schistosoma cercariae (150 卤2) escaped from Oncomelania dali and Oncomelania hupensis respectively. The serum total protein, albumin, globulin and alanine aminotransferase (ALT), in rabbits of control group and experimental group were detected by automatic biochemical analyzer at different time after infection.
