晚期糖基化终产物单克隆抗体的制备

【摘要】: 研究目的:为建立检测血清中晚期糖基化终末产物(AGEs)浓度的微量测定技术,为糖尿病慢性并发症的诊断、治疗及预后评估提供依据,本研究试图制备抗人AGEs单克隆抗体。 研究方法:D-葡萄糖和人血清白蛋白(HSA)溶解于磷酸盐缓冲液(PBS)中,37℃恒温孵育12周,同时以不含D-葡萄糖的HSA为对照。12周后透析凝胶过滤纯化,荧光测定、蛋白定量及聚丙烯酰胺凝胶电泳(SDS-PAGE)电泳鉴定。取100μg纯化后的第一峰AGE-HSA加等体积福氏佐剂免疫Babl/c小鼠,共3次,用聚乙二醇(PEG)行细胞融合,用杂交瘤细胞选择性培养基次黄嘌呤、氨基喋呤、胸腺嘧啶核苷(HAT)及次黄嘌呤、胸腺嘧啶核苷(HT)选择性培养,间接酶联免疫吸附实验(ELISA)筛选培养上清,包被的抗原为纯化后的第一峰AGE-HSA,选择对AGEs阳性,对HSA阴性的多克隆孔进行亚克隆,间接ELISA筛选亚克隆板孔的细胞培养上清。收集阳性单克隆细胞株,注射Babl/c小鼠制备腹水,经固相化甘露聚糖结合蛋白(MBP)柱纯化腹水。行亚型鉴定,并通过间接ELISA、western blot对抗体行特异性鉴定,并进行染色体鉴定。 结果:D-葡萄糖和HSA孵育12周后,反应液呈棕色,SDS-PAGE示分子量大于HSA的葡萄糖-HSA偶联产物,荧光测定表明产物系AGEs。经Sephadex G-200纯化后,得到高分子量和低分子量的两个蛋白峰,分别称为第一峰和第二峰。第一峰作为免疫原,免疫Babl/c小鼠后血清滴度大于1:106,细胞融合后筛选,获得阳性多克隆,经亚克隆,得到5株阳性单克隆细胞株。染色体鉴定示其染色体为106条。扩增5株细胞株,注射小鼠腹腔产生腹水,亚型鉴定为IgM型,抗体特异性鉴定示对AGE-HSA有特异性。 结论:获得抗人AGE-HSA单克隆抗体细胞株5株,抗体亚型为IgM型。
