新型乙肝表面抗原中蛋白核酸疫苗在中国猕猴体内免疫原性初步研究

【摘要】:目的:观察乙型肝炎病毒表面抗原中蛋白(MHBs)核酸疫苗在中国猕猴体内的体液和细胞免疫应答。方法:实验用中国猕猴实验组、对照组各2只,分别在第0、8、16、24、48周肌肉注射法接种核酸疫苗(pSW3891/MHBs)或对照载体(pSW3891)。ELISA法检测中国猕猴血清抗-HBs,CFSE-PI染色FCM(流式细胞术)法测定中国猕猴外周血淋巴细胞HBsAg特异性增殖反应。结果:实验组中国猕猴血清抗-HBs滴度在72周达到最高峰,最高滴度分别为1∶6400和1∶12800,对照组抗-HBs滴度始终保持在1∶50。实验组中国猕猴外周血淋巴细胞HBsAg特异性增殖反应明显增强,细胞分裂指数分别
