外周血有核细胞中tPA mRNA的表达与血栓性疾病的相关性研究

【摘要】:目的:血栓形成在许多心血管事件的发生中起着重要的作用,对人体健康危害极大。血栓性疾病的及时、准确诊断及溶栓治疗效果,对疾病的预后起着关键作用。组织型纤溶酶原激活物(t-PA)在人体纤溶系统中发挥着重要作用,其活性异常是心血管疾病血栓前状态的最危险因素。本实验采用RT-PCR法检测血栓形成组及血栓前状态组外周血有核细胞中t-PA mRNA的表达与正常人有无明显差异。 方法:收取了烟台毓璜顶医院神经内科及心血管内科自2004年10月至2004年12月收治的脑梗塞患者40例,采集外周血样本。同期取高血压患者20例,冠心病患者20例。正常对照40例,为健康查体者。分别用逆转录-PCR(RT-PCR)法检测其外周血有核细胞中t-PAmRNA的表达,发色底物法测定血浆t-PA的活性。 结果:血栓形成组及血栓前状态组外周血有核细胞中t-PA mRNA的表达均明显增强,同正常照组比较有显著性差异(P0.01),两组人群血浆中t-PA的活性也都明显降低,同正常对照组比较有显著性差异(P0.01)。外周血有核细胞中t-PA mRNA的表达与血栓前状态密切相关。实验结果采用均数±标准误表示,用Sigmastat 2.0统计软件进行统计学分析,所用统计方法有t检验、回归分析。P0.05,P0.01分别表示差别具有统计学意义和显著统计学意义。 结论:外周血有核细胞中t-PA mRNA的表达与血栓形成及血栓前状态密切相关,有可能作为血栓前状态的早期辅助监测指标。
[Abstract]:Objective: thrombosis plays an important role in the occurrence of many cardiovascular events and is very harmful to human health. The timely and accurate diagnosis and thrombolysis effect of thrombus diseases play a key role in the prognosis of thrombus diseases. Tissue type PA (t-PA) plays an important role in human fibrinolytic system, and its abnormal activity is the most risk factor for prethrombus state in cardiovascular diseases. In this study, RT-PCR assay was used to detect the expression of t-PA mRNA in peripheral blood nucleated cells in thrombus group and prethrombus state group, which was significantly different from that in normal subjects. Methods: from October 2004 to December 2004, 40 patients with cerebral infarction were collected from Department of Neurology and Cardiovascular Medicine of Yantai Yuhuangding Hospital. At the same time, 20 patients with hypertension and 20 patients with coronary heart disease were taken. 40 cases of normal control were examined for health. The expression of t-PAmRNA in peripheral blood nucleated cells was detected by reverse transcription-PCR (RT-PCR), and the activity of t-PA in plasma was measured by chromogenic substrate method. Results: the expression of t-PA mRNA in peripheral blood nucleated cells in thrombus group and prethrombus state group was significantly higher than that in normal control group (P 0.01). The activity of t-PA in plasma of the two groups was also significantly decreased. Compared with the normal control group, there was significant difference (P 0.01). The expression of t-PA mRNA in peripheral blood nucleated cells is closely related to prethrombus state. The experimental results were misrepresented by mean 卤standard, and statistically analyzed by Sigmastat 2.0 statistical software. The statistical methods used were t test and regression analysis. P05 and P01 showed that the difference was statistically significant and statistically significant. Conclusion: the expression of t-PA mRNA in peripheral blood nucleated cells is closely related to thrombosis and prethrombus state, and may be used as an early auxiliary monitoring index of prethrombus state.
