TGF-β1体外诱导人骨髓间充干细胞定向分化为成纤维细胞的研究

【摘要】: 研究目的:通过对照研究,用形态学,免疫组织化学方法分析TGF-β1在人骨髓间充质干细胞(bone marrow-derived Mesenchymal stem cells,骨髓MSCs)体外定向分化为成纤维细胞(fibroblast)过程中的作用,并比较TGF-β1在正常人和慢性骨髓增殖性疾病(chronic myeloproliferative disorders,CMPD)病人的骨髓间充质干细胞分化为成纤维细胞过程中的表达差异。方法:密度梯度离心法分离正常人和慢性骨髓增殖性疾病病人骨髓液中的间充质干细胞,用含10%胎牛血清(fetal calf serum,FCS)的DMEM低糖双抗培养液,,在37℃,5%CO_2培养箱中培养,0.25%胰蛋白酶+0.02%EDTA消化传代;5ng/ml的TGF-β1诱导分化骨髓MSCs,并作空白对照试验,分别用流式细胞分析细胞表面抗原,改良Masson三色染色法初步定性分析胶原蛋白的表达,免疫组织化学分析细胞基质蛋白属性。结果:联合应用密度梯度离心法和贴壁法能获得纯度较高,均一性较强的骨髓MSCs同源细胞,5ng/mlTGF-β1在骨髓间充质干细胞分化培养过程中,能促进MSCs表达胶原蛋白,波形蛋白。同等条件下,慢性骨髓增殖性疾病的MSCs胶原蛋白和波形蛋白表达高于正常人,细胞角蛋白的表达在各组中均低。讨论:通过密度梯度离心法结合贴壁法可以获得纯度较高的骨髓 间充质干细胞,呈梭形、三角形和多角形。表达CD29、CD44、CD166,不表达CD14、CD34、HLA-DR、CD45。在骨髓MSCs分化为成纤维细胞过程中,TGF-β1有促进分化的作用。同等条件下CMPD患者骨髓间充质干细胞较正常人的易于表达胶原蛋白和波形蛋白。
