仙台病毒HN基因核酸疫苗免疫幼鼠的效果评价

【摘要】:目的 人副流感病毒1型能够引起以哮吼为主要症状的呼吸道感染,是幼儿呼吸道感染而住院治疗的主要病原体之一,同时又可以累及成人。仙台病毒,又称鼠源副流感病毒1型,仙台病毒的HN糖蛋白与人源副流感病毒1型HN蛋白序列高度同源,并且是仙台病毒主要的抗原蛋白。因此,本研究用构建的仙台病毒HN核酸疫苗免疫幼鼠,评价疫苗的免疫效果,以寻求预防副流感病毒1型感染的新方法。 方法 用去内毒素处理后的仙台病毒HN核酸疫苗(pcDNA-HN)肌肉注射免疫4-6周龄幼鼠,加强免疫两次,4周后用仙台病毒攻击小鼠。以灭活的仙台病毒疫苗为阳性对照、pcDNA3和生理盐水作为阴性对照,采用1μg/0.1ml和5μg/0.1ml两个浓度的仙台病毒HN基因核酸疫苗做为观察对象。观察小鼠的肺组织病理学变化,通过B细胞和T细胞增殖反应、IFN-γ含量测定、血凝抑制试验、血清IgG抗体的测定及红细胞免疫功能测定等多项指标评价仙台病毒HN基因核酸疫苗的免疫应答效果。 结果 pcDNA-HN疫苗组肺指数显著低于生理盐水组、肺组织病理学无明显改变;与pcDNA3和生理盐水组比较,疫苗组可明显增强T、B淋巴细胞增殖能力,两者
[Abstract]:Objective Human parainfluenza virus type 1 can cause respiratory tract infection with roar as the main symptom. It is one of the main pathogens of respiratory tract infection in young children and can involve adults at the same time. Sendai virus, also known as mouse parainfluenza virus type 1, Sendai virus HN glycoprotein and human parainfluenza virus type 1 HN protein sequence is highly homologous, and is the main antigen protein of Sendai virus. Therefore, the constructed Sendai virus HN nucleic acid vaccine was used to immunize young mice and evaluate the immune effect of the vaccine in order to find a new method to prevent parainfluenza virus type 1 infection. Methods Sendai virus HN nucleic acid vaccine (pcDNA-HN) was intramuscular injected into young mice aged 4 weeks for 6 weeks and immunized twice. Four weeks later, the mice were attacked with Sendai virus (Sendai virus). The mice were immunized with Sendai virus (Sendai virus) intramuscular injection of Sendai virus nucleic acid vaccine (Sendai). The inactivated Sendai virus vaccine was used as positive control, pcDNA3 and saline as negative control, and 1 渭 g / 0.1ml and 5 渭 g / 0.1ml HN gene nucleic acid vaccine were used as observation objects. The pathological changes of lung tissue in mice were observed. the proliferation of B cells and T cells, the content of IFN- 纬 and the inhibition test of hemagglutination were measured. The determination of serum IgG antibody and erythrocyte immune function were used to evaluate the immune response effect of Sendai virus HN gene nucleic acid vaccine. Results the lung index of pcDNA-HN vaccine group was significantly lower than that of saline group, and there was no significant change in lung pathology. Compared with pcDNA3 and saline group, the proliferation ability of T and B lymphocytes in vaccine group was significantly enhanced.
