PDE4D基因多态性与支气管哮喘的相关性研究

【摘要】:目的研究磷酸二酯酶4D(PDE4D)基因rs1544791位点多态性与中国汉族儿童支气管哮喘的相关性。方法采用病例-对照的研究方法,选取2014年1月至2015年12月在青岛大学附属医院就诊的132名中国汉族支气管哮喘患儿及181名健康查体儿童作为研究对象,抽取外周静脉血,应用DNA提取试剂盒提取外周血白细胞DNA,应用聚合酶链反应(PCR)扩增,经外切核酸酶及虾碱酶纯化后,用连接酶反应连接荧光标记的特异探针,通过毛细管电泳来区分基因位点,并对比分析出两组人群的基因型频率和等位基因频率的差异,应用卡方检验对结果进行比较分析。结果哮喘组和对照组PDE4D基因rs1544791位点的分布均符合Hardy-Weinberg平衡定律,两组PDE4Drs1544791位点的3种基因型(TT、CT、CC)的分布及等位基因频率与正常对照组相比差异均无统计学意义(p0.05)。结论磷酸二酯酶4D(PDE4D)基因rs1544791位点多态性与中国汉族儿童支气管哮喘无相关性。
[Abstract]:Objective to study the association between the rs1544791 polymorphism of phosphodiesterase 4D (PDE4D) gene and bronchial asthma in Chinese Han children. From January 2014 to December 2015, 132 children with bronchial asthma of Han nationality and 181 healthy children were selected as study subjects, and peripheral venous blood was drawn. The peripheral blood leukocyte DNA was extracted by DNA extraction kit and amplified by polymerase chain reaction (PCR). After purified by exonuclease and shrimp base enzyme, ligase reaction was used to connect the specific probe labeled with fluorescence, and the loci were identified by capillary electrophoresis. The differences of genotype frequency and allele frequency between the two groups were compared and analyzed by chi-square test. Results the distribution of rs1544791 loci of PDE4D gene in asthmatic group and control group was in accordance with the law of Hardy-Weinberg equilibrium. There was no significant difference in the distribution and allelic frequency of the three genotypes of PDE4Drs1544791 between the two groups compared with the control group (P 0.05). Conclusion there is no correlation between the polymorphism of rs1544791 locus of phosphodiesterase 4DG PDE4D gene and bronchial asthma in Chinese Han children.
