婴幼儿血管瘤组织中β2受体的表达分布及意义

【摘要】:目的:通过免疫荧光技术检测β2受体在婴幼儿血管瘤组织中的表达及分布特点,探讨其在血管瘤组织病理演变过程中的作用及意义 方法:将48例婴幼儿血管瘤进行HE染色,,根据临床表现和组织学特征,分为增殖期婴幼儿血管瘤组和消退期婴幼儿血管瘤组,其中增殖期婴幼儿血管瘤29例,消退期婴幼儿血管瘤19例。采用免疫荧光技术分别将两组婴幼儿血管瘤组织中内皮细胞核及β2受体进行荧光标记,观察两组照片荧光标记物的显示位置,并应用Image-ProPlus6.0软件先将荧光照片转换为黑白图片然后对每张照片进行分析,得出内皮细胞核及β2受体阳性的平均光密度值。 结果:增殖期婴幼儿血管瘤组内皮细胞的平均光密度值为0.031±0.002,消退期婴幼儿血管瘤组β2受体的平均光密度值均数为0.022±0.002,两者具有显著统计学差异。增殖期婴幼儿血管瘤组β2受体的平均光密度值为0.034±0.003,消退期婴幼儿血管瘤组β2受体的平均光密度值均数为0.027±0.002,两者具有显著统计学差异。增殖期婴幼儿血管瘤中β2受体的表达明显高于消退期婴幼儿血管瘤。荧光图片显示β2受体于婴幼儿血管瘤中广泛表达,并以内皮细胞核荧光表达处为主。 结论:增殖期婴幼儿血管瘤中内皮细胞及β2受体的表达均明显高于消退期婴幼儿血管瘤,消退期婴幼儿血管瘤组织中内皮细胞核的消退伴随β2受体表达的减弱,可推测β2受体主要广泛表达于婴幼儿血管瘤内皮细胞中,且可能存在于周细胞中。提示β2受体阻滞剂剂作用于婴幼儿血管瘤,诱导其消退的机制可能与内皮细胞及周细胞中β2受体有关。
[Abstract]:Objective: to detect the expression and distribution of 尾 2 receptor in infantile hemangioma by immunofluorescence technique. Methods: 48 cases of infantile hemangioma were stained with HE according to their clinical manifestations and histological features. There were 29 cases of proliferative hemangioma and 19 cases of involutive hemangioma in infantile hemangioma. The endothelial nuclei and 尾 2 receptors were labeled with immunofluorescence technique in two groups of infantile hemangioma tissues, and the display position of the fluorescent markers in the two groups were observed. Using Image-ProPlus6.0 software, the fluorescence photos were converted into black and white pictures, and then each photo was analyzed, and the average optical density of endothelial cell nucleus and 尾 2 receptor positive was obtained. Results: the average optical density of endothelial cells was 0.031 卤0.002 in proliferative hemangioma group and 0.022 卤0.002 in involutive phase. The average optical density of 尾 2 receptor was 0.034 卤0.003 in proliferative hemangioma group and 0.027 卤0.002 in involutive phase. There was significant difference between the two groups. The expression of 尾 2 receptor in proliferative hemangioma was significantly higher than that in involutive hemangioma. Fluorescence images showed that 尾 2 receptor was widely expressed in infantile hemangioma, and mainly in endothelial cell nucleus. Conclusion: the expression of endothelial cells and 尾 2 receptor in proliferative hemangioma is significantly higher than that in involutive hemangioma. It can be inferred that 尾 2 receptor is mainly expressed in endothelial cells of infantile hemangioma and may exist in pericytes. It is suggested that the mechanism of 尾 2 receptor antagonist acting on infantile hemangioma may be related to 尾 2 receptor in endothelial cells and pericytes.
