兔实验性大肠埃希菌脑膜炎脉络丛GLUT1的变化

【摘要】:目的: 在兔实验性大肠埃希菌脑膜炎中观察兔一般情况、脑脊液改变,初步探讨细菌性脑膜炎时是否存在脉络丛组织上GLUT1蛋白受损,是否与脑脊液糖/血糖比值的变化有关。 方法: 1.验证兔脉络丛组织GLUT1的表达 2.制备兔实验性大肠埃希茵脑膜炎模型。 3.观察家兔一般情况,小脑延髓池穿刺取脑脊液进行相关检测,并对各指标进行相关性分析。 4.采用Real-time PCR和West-blot检测脑脉络丛组织GLUT1基因mRNA水平和蛋白水平的表达。 结果: 1.本次实验验证了脉络丛组织中存在GLUT1的表达 2.本研究成功制备了家兔大肠埃希菌脑膜炎动物模型。 3.与注菌前及对照组比较:模型组家兔体温升高(P0.05),同时伴随精神、进食、活动等改变;脑脊液外观浑浊,白细胞数、蛋白含量明显升高(P0.01),脑脊液乳酸升高(P0.01),脑脊液糖、脑脊液糖/血糖比值明显下降(P0.01),差异均有统计学意义,脑脊液白细胞数、蛋白与脑脊液乳酸、脑脊液糖及脑脊液糖/血糖比值各指标间比较有相关性。 4.模型组的脉络丛组织GLUT1mRNA和蛋白相对表达量减少(P0.05)。 结论: 1兔实验性大肠埃希菌脑膜炎时脑脊液乳酸显著升高,脑脊液糖含量及脑脊液糖/血糖比值有明显降低,且脑脊液糖/血糖比值小于0.5,且以上各项指标与脑脊液白细胞数、脑脊液蛋白具有相关性。 2.本研究首次提出细菌性脑膜炎时脉络丛组织上GLUT1蛋白受损,可能与脑脊液糖/血糖比值下降有关。
[Abstract]:Objective: to observe the general situation and changes of cerebrospinal fluid (CSF) in rabbits with experimental Escherichia coli meningitis, and to explore whether GLUT1 protein was damaged in choroid plexus tissue during bacterial meningitis. Whether or not it is related to the change of CSF sugar / glucose ratio. Methods: 1. To verify the expression of GLUT1 in choroidal plexus of rabbits. A rabbit model of experimental Escherichia coli meningitis was established. Observe the general situation of rabbits, cerebellar medullary cistern puncture to take cerebrospinal fluid for correlation detection, and the correlation analysis of each index. 4. 4. Real-time PCR and West-blot were used to detect the expression of GLUT1 mRNA and protein in choroid plexus. Results: 1. This experiment confirmed the existence of GLUT1 expression in choroidal plexus. The animal model of Escherichia coli meningitis in rabbits was established successfully. Compared with the control group and before the injection of bacteria, the temperature of the model group increased (P0.05), accompanied by mental, eating, activity and other changes, cerebrospinal fluid appearance turbidity, white blood cell count, protein content significantly increased (P0.01), cerebrospinal fluid lactic acid (P0.01), cerebrospinal fluid sugar, and so on, The ratio of CSF sugar / glucose decreased significantly (P0.01). The number of CSF leukocytes and protein were correlated with cerebrospinal fluid lactic acid, CSF sugar and CSF sugar / glucose ratio. 4. The relative expression of GLUT1 mRNA and protein in choroid plexus was decreased in model group (P0.05). Conclusion: 1 the level of cerebrospinal fluid (CSF) lactic acid and the ratio of cerebrospinal fluid sugar (CSF) to glucose (CSF) were significantly decreased in rabbits with experimental Escherichia coli meningitis. The ratio of CSF sugar to glucose was less than 0.5, and the above indexes were correlated with the number of CSF leukocytes and cerebrospinal fluid (CSF) protein. 2. This study suggested for the first time that GLUT1 protein in choroid plexus was damaged in bacterial meningitis, which may be related to the decrease of CSF glucose / glucose ratio.
