ICAM-1、MCP-1表达水平及基因多态性与EV71手足口病

【摘要】:目的研究ICAM-1基因K469E位点、MCP-1A2518G位点基因多态性及sICAM-1、MCP-1在血清中表达水平与EV71手足口病的关系,探讨EV71型手足口病的遗传易感因素。方法收集手足口病患儿急性期(发病24小时内)大便标本,常规处理大便标本,运用RT-PCR方法,采用肠道病毒71型(EV71)特异引物扩增检测病毒RNA。采用双夹心抗体法(ELISA)检测EV71感染阳性组和对照组血清sICAM-1和MCP-1水平,并进行直线相关分析。运用限制性片段长度多态性—聚合酶链反应(PCR-RFLP)检测EV71感染阳性组和对照组中ICAM-1K469E位点及MCP-1A2518G位点碱基变异情况,并取样采用碱基直接测序进行验证,对数据进行统计学分析。结果①306例手足口病患儿大便标本检测中153例EV71阳性,其中男83例,女70例,年龄11月~6岁,平均年龄2.5岁。②手足口病组患儿血清中sICAM-1和MCP-1水平均显著高于正常对照组(P均0.01)。③EV71手足口病组和对照组ICAM-1K469E位点基因型分布存在显著差异(x2=12.433,P0.01),EV71手足口病组A等位基因频率明显低于对照组(x2=8.073,P0.01)。④EV71手足口病组患儿MCP-1基因型分布、等位基因频率与对照组比较均无统计学意义(P0.05)。结论①ICAM-1和MCP-1表达水平与EV71手足口病有关,推测sICAM-1参与机体EV71感染后的炎症反应和免疫损伤过程。②ICAM-1基因K469E位点多态性与EV71手足口病相关,推测A等位基因是EV71感染的保护性基因,可降低EV71手足口病发生率。③MCP-1基因A-2518G位点基因多态性与EV71手足口病无关。
[Abstract]:Objective to study the relationship between the polymorphism of ICAM-1 gene K469E locus MCP-1A2518G and the expression level of sICAM-1 MCP-1 in serum with EV71 hand-foot-mouth disease (HFMD), and to explore the genetic susceptibility factors of EV71 Hand-foot-mouth disease (HMD). Methods stool specimens of children with hand-foot-mouth disease (HFMD) in acute phase (within 24 hours of onset) were collected. Stool specimens were treated with routine method. RT-PCR method was used to detect the viral RNA by using specific primers of enterovirus 71 (EV71). The serum levels of sICAM-1 and MCP-1 were detected by double sandwich antibody (ELISA) in the positive group of EV71 infection and the control group, and the linear correlation analysis was carried out. Restriction fragment length polymorphism (PCR-RFLP) was used to detect the variation of ICAM-1K469E and MCP-1A2518G loci in EV71 positive group and control group. The data were analyzed by direct sequencing. Results among the 1306 stool specimens of hand, foot and mouth disease, 83 were male and 70 were female. The age was 6 years old in November. The mean age of 2.5 years old in the HFMD group was significantly higher than that in the normal control group (P0.01). 3There was a significant difference in the distribution of ICAM-1K469E genotypes between the HFMD group and the control group (x2v12.433P0.01). The frequency of MCP-1 gene was significantly lower than that of control group (x2 + 8.073U, P0.01) .4EV71 Hand-foot-mouth disease (HFMD) group, the distribution of MCP-1 genotype was significantly lower than that in control group. There was no significant difference in allele frequency between the control group and the control group (P0.05). Conclusion the expression of 1ICAM-1 and MCP-1 is related to EV71 HFMD. It is speculated that sICAM-1 is involved in the inflammatory reaction and the process of immune damage after EV71 infection. The K469E polymorphism of K469E gene of ICAM-1 gene is associated with EV71 HFMD. It is suggested that A allele is a protective gene for EV71 infection, which can reduce the incidence of EV71 HFMD. 3The polymorphism of A-2518G locus of MCP-1 gene is not related to EV71 HFMD.
