针刺对缺氧新生鼠脑NeuroD表达的影响

【摘要】:【目的】 建立新生鼠脑缺氧模型,模拟临床新生儿缺氧缺血性脑病(HIE),通过对模型鼠电针刺激处理后,研究针刺对缺氧新生鼠脑中神经源性分化因子(NeuroD)表达及细胞增殖的影响,为针刺治疗HIE疾病提供实验依据。 【方法】 1.按照Grojean的方法:将出生10-24h内SD新生大鼠置于100%氮气的无氧环境中,建立HIE模型,并对所做的动物模型进行评价。 2.将动物随机分为三组:对照组、缺氧组、缺氧+电针刺组,电针组采用临床所用电子治疗仪器,,选取大鼠相应的穴位,每天持续30min,连续针刺10d后,通过RT-PCR、western blot及免疫荧光法检测NeuroD,免疫荧光法检测5-溴脱氧尿嘧啶核苷(BrdU)表达的变化。 【结果】 1.缺氧过程中新生大鼠均出现神经异常的表现,如:躁动、意识障碍,并伴有全身紫绀,大脑皮质HE染色可见缺氧组鼠脑神经细胞损伤。 2.免疫荧光示:神经干细胞聚集区大脑皮质、海马、SVZ等部位有NeuroD的表达。RT-PCR结果示:针刺组NeuroD mRNA表达量高于均正常组和缺氧组(P<0.01)。Western blot结果示:针刺组NeuroD/β-actin平均光密度均高于正常组和缺氧组(P<0.01)。针刺组NeuroD蛋白平均荧光强度均高于正常组和缺氧组(P<0.01);针刺组SVZ、DG部位BrdU表达量均高于缺氧组和对照组(P<0.01)。 【结论】 电针刺激能使鼠脑神经干细胞聚集区的大脑皮质、海马、SVZ等部NeuroD表达量增高,并可诱导细胞增殖,可能参与受损神经组织修复和功能重建。
[Abstract]:[objective] to establish a neonatal model of hypoxic brain, and to simulate the clinical hypoxic-ischemic encephalopathy (HIE),) of neonatal hypoxic-ischemic encephalopathy (HIE). To study the effect of acupuncture on the expression of neurogenic differentiation factor (NeuroD) and cell proliferation in the brain of hypoxic neonatal rats, and to provide experimental evidence for acupuncture treatment of HIE disease. [methods] 1. According to the method of Grojean, the neonatal rats of SD were placed in 100% nitrogen anaerobic environment within 10-24 hours. The HIE model was established, and the animal model was evaluated. 2. The animals were randomly divided into three groups: control group, anoxic electroacupuncture group, electroacupuncture group and electroacupuncture group. The rats were selected corresponding acupoints for 30 minutes every day, after 10 days of continuous acupuncture, RT-PCR, was adopted. The expression of 5-bromodeoxyuridine (5-bromodeoxyuridine) (BrdU) was detected by western blot and NeuroD, immunofluorescence assay. [result] 1. During anoxia, the neonatal rats showed abnormal neurological manifestations, such as restlessness, disturbance of consciousness, and cyanosis. HE staining of cerebral cortex showed brain nerve cell damage in hypoxic group. 2. Immunofluorescence: neural stem cell accumulation, cerebral cortex, hippocampus, The expression of NeuroD was found in SVZ and other parts. RT-PCR results showed that the expression of NeuroD mRNA in acupuncture group was higher than that in normal group and hypoxia group (P < 0. 01). Western blot): the average optical density of NeuroD/ 尾-actin in acupuncture group was higher than that in normal group and deficient group (P < 0. 01). Oxygen group (P < 0.01). The average fluorescence intensity of NeuroD protein in acupuncture group was higher than that in normal group and hypoxia group (P < 0. 01), and the expression of BrdU in SVZ,DG in acupuncture group was higher than that in hypoxia group and control group (P < 0. 01). [conclusion] electroacupuncture can increase the expression of NeuroD in cerebral cortex, hippocampus and SVZ, and induce cell proliferation, which may be involved in the repair and functional reconstruction of injured nerve tissue.
