维生素E及叶酸对高血糖孕鼠子代心肌凋亡的影响

【摘要】:目的:探讨维生素E(Ve)及叶酸对高血糖诱导的乳鼠心肌细胞凋亡蛋白capase-3表达的影响。方法:将50只清洁级成年SD孕鼠随机分为5组,分别为正常对照组(A组)、妊娠糖尿病组(B组)、叶酸组(C组)、维生素E组(D组)和Ve+叶酸组(E组)。除对照组外,其余大鼠于妊娠第3天腹腔注射链脲佐菌素(STZ)35mg/kg制备妊娠糖尿病动物模型,对照组给予等量0.1mmol/L(PH4.5)柠檬酸钠—柠檬酸缓冲液。孕第5天,VE组、VE+叶酸组两组给予50mg/kg维生素E灌胃,叶酸组、VE+叶酸组予0.4mg/kg叶酸灌胃,A、B组予等量的生理盐水灌胃,持续7天。取自然分娩的乳鼠心肌组织于光镜下观察心肌结构的改变,应用免疫组织化学法检测各组乳鼠心肌细胞caspase-3抗原的表达。蛋白免疫印迹(western blot)检测各组乳鼠心肌细胞凋caspase-3的表达。结果:1.造模前各组血糖值比较差异无统计学意义(p0.05);造模后,糖尿病、叶酸组、VE组、VE+叶酸组血糖值明显高于A组,差异有统计学意义(p0.01);糖尿病、叶酸组、VE组、VE+叶酸组四组间比较差异无统计学意义(p0.05)。2.光镜下观察糖尿病、叶酸组、VE组、VE+叶酸组乳鼠心肌组织呈现不同程度损伤,表现为细胞坏死、水肿、轮廓不清,心肌纤维排列紊乱、炎性细胞浸润。3.5组间乳鼠心肌细胞caspase-3平均吸光度值两两比较,差异有统计学意义(p0.01)。4.5组胎鼠心肌细胞caspase-3蛋白表达灰度值两两比较,差异有统计学意义(p0.01)结论:孕鼠妊娠期高血糖可导致胎鼠心肌细胞凋亡增多,补充VE和叶酸对妊娠高血糖所致的胎儿心肌细胞凋亡有保护作用,叶酸抑制胎鼠心肌凋亡的作用强于VE,,联合补充VE和叶酸较单纯补充VE或叶酸效果更佳,但不能使其恢复正常水平。
[Abstract]:Objective: to investigate the effects of Vitamin E (Veg) and folic acid on the expression of apoptotic protein (capase-3) in neonatal rat cardiomyocytes induced by hyperglycemia. Methods: fifty adult SD pregnant rats of clean grade were randomly divided into 5 groups. They were normal control group A, gestational diabetes mellitus group B, folic acid group C group, vitamin E group D) and ve folic acid group. The other rats were intraperitoneally injected with STZ 35 mg / kg on the third day of pregnancy to make gestational diabetic animal model, the control group was given the same amount of 0.1 mmol / L PH4.5) citrate sodium citric acid buffer, and on the 5th day of pregnancy, the VE folic acid group and the VE folic acid group were given 50 mg / kg vitamin E intragastrically. The rats in folic acid group were given 0.4 mg / kg folic acid intragastric perfusion with normal saline for 7 days. The myocardial structure of naturally delivered neonatal rats was observed under light microscope. Immunohistochemical method was used to detect the expression of caspase-3 antigen in neonatal rat cardiomyocytes. Western blotting was used to detect the expression of caspase-3 in neonatal rat cardiomyocytes. Results: 1. There was no significant difference in blood glucose levels before and after modeling. In diabetes mellitus, the blood glucose level in VE group was significantly higher than that in A group, the difference was statistically significant (p 0.01), and there was no significant difference among the four groups in diabetes mellitus (DM) and folic acid group (VE). In folic acid group, VE group showed different degree of injury in myocardial tissue, including cell necrosis, edema, unclear outline, disordered arrangement of myocardial fibers, inflammatory cell infiltration. The average caspase-3 absorbance of myocardial cells of neonatal rats in 3.5 groups was compared in pairs. There was a significant difference in the expression of caspase-3 protein in fetal cardiomyocytes between the two groups (P < 0.01). Conclusion: hyperglycemia during pregnancy can lead to the increase of cardiomyocyte apoptosis in fetal rats. Vitamin E and folic acid supplementation had protective effect on fetal cardiomyocyte apoptosis induced by pregnancy hyperglycemia. Folic acid had stronger effect on inhibiting fetal myocardial apoptosis than VE.The combination of VE and folic acid was better than vitamin E or folic acid alone. But it cannot be brought back to normal.
