苦参碱及联合顺铂对宫颈癌SiHa细胞中TSLC1基因表达的影响

【摘要】:目的探讨苦参碱及联合顺铂对宫颈癌Si Ha细胞中TSLC1基因表达的影响。方法以人宫颈癌细胞株Si Ha细胞株为试验材料,采取甲基偶氮唑蓝法检测并计算细胞抑制率。采取实时荧光逆转录聚合酶联反应法分析TSLC1 mRNA基因表达情况。结果顺铂、苦参碱联合顺铂均可显著抑制Si Ha细胞体外增殖,差异具有统计学意义(P0.05);随着用药浓度的增加,抑制作用增强,不同浓度两两比较,差异具有统计学意义(P0.05);联合组细胞抑制率高于苦参组、顺铂组,差异具有统计学意义(P0.05)。三组用药后,TSLC1基因表达水平均高于对照组,差异具有统计学意义(P0.05);随着浓度的增加,影响效果更强,组间不同浓度两两比较,差异具有统计学意义(P0.05);联合组高于顺铂组、苦参碱组,同浓度比较,差异具有统计学意义(P0.05);顺铂组与苦参碱组同浓度比较,差异无统计学意义(P0.05)。结论苦参碱及联合顺铂对宫颈癌Si Ha细胞中TSLC1基因表达有抑制作用,且与药物浓度呈剂量依赖性。
[Abstract]:Objective to investigate the effect of matrine and cisplatin on the expression of TSLC1 gene in cervical cancer Si Ha cells. Methods Human cervical cancer cell line Si Ha cell line was used as the experimental material, and the inhibition rate was measured by methyl azolium blue method. The expression of TSLC1 mRNA gene was analyzed by real-time fluorescence reverse transcriptase polymerase chain reaction (RT-PCR). Results Cisplatin, matrine and cisplatin could significantly inhibit the proliferation of Si Ha cells in vitro (P0.05). The difference was statistically significant (P0.05); the cell inhibition rate of combined group was higher than that of Sophora flavescens group, cisplatin group, the difference was statistically significant (P0.05). The expression level of TSLC1 gene in the three groups was significantly higher than that in the control group (P0.05); with the increase of the concentration, the effect was stronger, the difference was statistically significant (P0.05); the combined group was higher than the cisplatin group (P0.05). Matrine group, the same concentration comparison, the difference was statistically significant (P0.05); cisplatin group and matrine group with the same concentration comparison, the difference was not statistically significant (P0.05). Conclusion Matrine and cisplatin can inhibit the expression of TSLC1 gene in cervical cancer Si Ha cells in a dose-dependent manner.
