【摘要】:【目的】探讨超顺磁性氧化铁(SPIO)标记大鼠脂肪间充质干细胞(RADSC)后,其成骨样分化细胞中铁调节蛋白1(IRP1)基因及蛋白表达的变化。【方法】本实验采用转染剂多聚赖氨酸(PLL)介导SPIO标记RADSC,然后将其诱导为成骨样分化细胞,分别在诱导后0、7、14、21 d进行RT-PCR及Western blot检测其IRP1基因及蛋白的表达。【结果】成骨样分化细胞IRP1 mRNA表达水平在SPIO标记与未标记组之间差异均无统计学显著性意义(F=1.571,P=0.239);IRP1蛋白表达水平在标记与未标记组之间差异均无统计学显著性意义(F=0.547,P=0.500)。【结论】SPIO标记并未对成骨样分化细胞的IRP1基因及IRP1蛋白表达产生明显影响。
[Abstract]:[objective] to investigate the labeling of rat adipose mesenchymal stem cells (RADSCs) by superparamagnetic iron oxide (SPIO). The changes of iron regulatory protein 1 (IRP1) gene and protein expression in osteoblast-like differentiation cells. [methods] SPIO labeled RADSC was mediated by polylysine transducer (PLL). Then they were induced into osteoblast-like differentiation cells, respectively. RT-PCR and Western blot were used to detect the expression of IRP1 gene and protein in 21 days. [results] Osteoblast-like differentiation cell IRP1. There was no significant difference in the expression of mRNA between SPIO labeled and unlabeled groups (P < 0.05). FN 1.571. Pu 0.239; There was no significant difference in the expression of IRP1 protein between labeled and unlabeled groups. [conclusion] SPIO labeling has no significant effect on the expression of IRP1 gene and IRP1 protein in osteoblast-like differentiation cells.
