高频电刺激脚桥核对苍白球内侧部氨基酸类递质及神经元放电的影响

【摘要】:目的通过观察高频电刺激帕金森(PD)模型大鼠脚桥核(PPN)对苍白球内侧部(GPi)氨基酸类递质及神经元放电的影响,探讨电刺激PPN治疗PD的机制。方法将70只SD雄性大鼠随机分为对照组30只,PD模型组40只。大脑右侧黑质致密部两点注射6-羟基多巴胺建立PD模型,应用细胞外单位记录观察高频电刺激PPN对GPi神经元放电的影响。脑内微透析收集损毁侧GPi脑脊液,结合高效液相色谱检测氨基酸类递质的变化。结果高频电刺激PPN,对照组及PD模型组大鼠GPi神经元的反应均以兴奋性为主,且平均放电频率均较刺激前明显增高(P㩳0.01);高频电刺激PPN,GPi内谷氨酸水平较刺激前明显减少(P㩳0.01),γ-氨基丁酸水平较刺激前无明显变化(P㧐0.05)。结论高频电刺激PPN可增强GPi神经元的电活动,降低GPi内谷氨酸的水平,其兴奋GPi神经元的效应可能是通过PPN-GPi直接通路或其他间接通路实现的。
[Abstract]:Objective to observe the effects of high frequency electrical stimulation on the amino acid transmitters and neuronal discharges of GPiG in the medial part of globus pallidus. Methods 70 male SD rats were randomly divided into control group (n = 30) and control group (n = 30). PD model group (n = 40). PD model was established by two points injection of 6-hydroxydopamine into the right substantia nigra compact part of the brain. The effects of high frequency electrical stimulation (PPN) on the discharge of GPi neurons were observed by extracellular unit recording. Cerebrospinal fluid (CSF) was collected from the damaged side of GPi by intracerebral microdialysis. Results the responses of GPi neurons to high frequency electrical stimulation, control group and PD model group were mainly excitatory. The average discharge frequency was significantly higher than that before stimulation. Glutamic acid levels in PPNG GPI were significantly reduced by high frequency electrical stimulation compared with those before stimulation. The levels of 纬 -aminobutyric acid were not significantly changed compared with those before stimulation. Conclusion High frequency electrical stimulation of PPN can enhance the electrical activity of GPi neurons and decrease the level of glutamate in GPi. The effect of stimulating GPi neurons may be realized through PPN-GPi direct pathway or other indirect pathway.
