苏木乙酸乙酯提取物对动脉粥样硬化模型大鼠血清IL-18、IL-10的影响

【摘要】:目的:通过建立动脉粥样硬化模型大鼠,应用苏木乙酸乙酯提取物干预,观察大鼠腹主动脉血管壁病理形态学改变及大鼠血清IL-18、IL-10水平及IL-18/IL-10变化,为苏木乙酸乙酯提取物抗动脉粥样硬化作用提供客观依据。方法:将60只SD大鼠随机分为6组,分别为:空白对照组、模型对照组、苏木乙酸乙酯提取物低剂量组、苏木乙酸乙酯提取物中剂量组、苏木乙酸乙酯提取物高剂量组、辛伐他汀组。空白对照组给予普通饲料喂养,其余5组行腹腔注射维生素D3+高脂饲料喂养进行动脉粥样硬化模型制备。模型复制成功后,给予空白对照组、模型对照组10mg/ml等体积羧甲基纤维素钠溶液,辛伐他汀组予10mg/ml等体积辛伐他汀溶液,相当于辛伐他汀剂量4.2mg/(kg·d),苏木乙酸乙酯提取物低、中、高剂量组给予10mg/ml等体积低、中、高浓度的苏木乙酸乙酯提取物溶液,分别相当于生药量0.0575mg/(kg.d)、0.115mg/(kg·d)、0.230mg/(kg.d),每日给药 1 次,连续给药4周。应用HE染色法观察各组大鼠腹主动脉病理形态学改变,酶联免疫吸附法检测各组大鼠血清IL-18、IL-10含量变化。结果:1.与模型对照组相比,辛伐他汀组及苏木乙酸乙酯提取物高、中、低剂量组大鼠腹主动脉血管壁的病理损伤程度减轻。2.与空白对照组相比,模型对照组、辛伐他汀组、苏木乙酸乙酯提取物高、中、低剂量组血清IL-18水平升高,差异有统计学意义(P0.05);与空白对照组相比,模型对照组、辛伐他汀组及苏木乙酸乙酯提取物高、中、低剂量组血清IL-10水平升高,差异有统计学意义(P0.05)。3.与模型对照组相比,辛伐他汀组及苏木乙酸乙酯提取物高、中、低剂量组血清IL-18水平降低,血清IL-10水平升高,差异有统计学意义(P0.05)4.与辛伐他汀组相比,苏木乙酸乙酯提取物高、中、低剂量组血清IL-18水平及IL-10水平无差异(P0.05)。结论:1.苏木乙酸乙酯提取物能够减轻动脉粥样硬化模型大鼠腹主动脉血管壁的病理损伤。2.苏木乙酸乙酯提取物能够降低动脉粥样硬化模型大鼠血清IL-18水平,升高血清IL-10水平,从而使血清IL-18/IL-10趋于平衡,维持机体致炎/抑炎平衡,降低机体炎症反应活性。
[Abstract]:Objective: to observe the pathomorphological changes of abdominal aorta vascular wall and serum IL-18 of rats by using ethyl acetate extract of hematoxylum to observe the pathomorphology of abdominal aorta in rats. The level of IL-10 and the change of IL-18/IL-10 provided the objective basis for the anti-atherosclerosis effect of ethyl acetate extract. Methods: 60 Sprague-Dawley rats were randomly divided into 6 groups. They were blank control group, model control group, low dose group, middle dose group and high dose group. Simvastatin group. The blank control group was fed with common diet, the other five groups were fed with vitamin D3 high fat diet to prepare the atherosclerosis model. After the model was successfully replicated, the blank control group was given. The model control group was given 10mg / ml sodium carboxymethyl cellulose solution, and the simvastatin group was given 10mg / ml simvastatin solution of the same volume, which was equivalent to the simvastatin dosage of 4.2 mg / kg 路d). The ethyl acetate extract of Sappa chinensis was low, medium and high dose group was given 10mg / ml solution of ethyl acetate extract of low, medium and high concentration. The same amount as 0.0575 mg / kg / kg / kg / kg / kg / d, 0.230 mg / r / kg / kg / d, respectively, was given once a day. The pathomorphology of abdominal aorta of rats in each group was observed by HE staining and serum IL-18 was detected by enzyme-linked immunosorbent assay (Elisa). Results: 1. Compared with the model control group, simvastatin group and hematoxylum ethylacetate extract were higher than those in the model control group. Compared with the blank control group, the model control group, simvastatin group, ethyl sappan acetate extract was high, medium. The serum IL-18 level in the low dose group was higher than that in the control group (P 0.05). Compared with the blank control group, the level of serum IL-10 in simvastatin group, simvastatin group and ethyl acetate group was higher than that in control group, while the level of serum IL-10 in medium and low dose groups was higher than that in control group. Compared with the model control group, simvastatin group and hematoxylum acetate ethyl acetate extract were higher, and the serum IL-18 level was lower in medium and low dose groups. The level of serum IL-10 was higher than that of simvastatin group, the difference was statistically significant (P 0.05). Compared with simvastatin group, the extract of ethyl acetate of hematoxylum was higher than that of simvastatin group. There was no difference in serum IL-18 and IL-10 levels in low dose group (P0.05). ... conclusion:. 1. Ethyl Sappa acetate extract can reduce the pathological damage of abdominal aorta vascular wall in atherosclerotic rats. 2. Ethyl Acetate extract of Sappa acetate can reduce serum IL-18 in atherosclerotic rats. Level. The level of serum IL-10 was increased, so that serum IL-18/IL-10 tended to balance, maintain the balance of inflammation and inhibition of inflammation, and reduce the activity of inflammatory reaction.
