狂犬病毒核蛋白时间分辨免疫荧光分析试剂盒研制

【摘要】：目的研制狂犬病毒(RV)核蛋白(NP)时间分辨免疫荧光分析(TRFIA)试剂盒。方法采用双抗体夹心法建立RV NP TRFIA试剂盒,对试剂盒的各项指标进行评价。结果试剂盒准确度好,线性范围为5～2 500mEU/mL,灵敏度为1.018mEU/mL,精密度良好,分析内精密度为2.7%～9.3%,分析间精密度为3.5%～12.2%。将15份疫苗样品用本法与国内常用ELISA检测试剂盒同时检测,其相关系数r为0.85,稳定性试验表明试剂可以在4℃稳定半年,37℃稳定7d。结论试剂盒各项已检测指标达到临床检验要求,有望进一步作临床检测试验。
[Abstract]:Objective to develop a time resolved immunofluorescence assay (TRFIA) kit for nucleoprotein NPV of rabies virus (RV). Methods the RV NP TRFIA kit was established by double antibody sandwich method, and the indexes of the kit were evaluated. The calibration curve was linear in the range of 5 ~ 2 500 mEU / mL, with a sensitivity of 1.018 mEU-mL. the precision was good, the analytical precision was 2. 7% and 9. 3%, and the analytical precision was 3. 5% and 12. 2%. 15 samples of vaccine were detected by this method and the ELISA kit used in our country. The correlation coefficient r is 0.85. The stability test shows that the reagent can be stabilized at 4 鈩，
