尖叶假龙胆对佐剂型关节炎模型大鼠的研究

【摘要】:目的:以尖叶假龙胆为研究对象,通过尖叶假龙胆对佐剂型关节炎模型大鼠的药效学实验。方法:以佐剂性关节炎模型大鼠,将SD大鼠随机分为模型组、空白组、阳性组(雷公藤多苷)、尖叶假龙胆高剂量组、尖叶假龙胆中剂量组、尖叶假龙胆低剂量组,每组8只。除空白组外在其余大鼠右后足足跖皮下注射弗氏完全佐剂(CFA)0.1ml致炎,复制模拟AA模型,空白组用生理盐水同法注射。从第14天开始灌胃,连续灌胃18天。定期测量大鼠体重,右足足趾肿胀度,踝关节和脾脏病理HE,ELISA法检测大鼠血清中的TNF-α、IL-10含量,免疫组化法检测滑膜组织里的VEGF、ICAM-1、MMP-3蛋白表达。结果:1尖叶假龙胆各剂量组可以增加AA大鼠的体重、减少AA大鼠足趾的肿胀、降低AA大鼠关节炎评分、降低AA大鼠脾脏白髓的生成。2.尖叶假龙胆各剂量组可以通过降低AA大鼠血清中的TNF-α的含量及提升血清IL-10的含量,以达到缓解佐剂性关节炎的作用。(P0.05)3.尖叶假龙胆各剂量组可以降低AA大鼠滑膜组织VEGF、ICAM-1、MMP3蛋白表达,从而达到缓解滑膜炎症、减轻关节肿胀、延缓关节骨破坏的目的。其中高剂量组效果最明显。(P0.05,P0.01)结论:尖叶假龙胆能有效的抑制继发性关节肿胀,可显著降低AA大鼠血清中的TNF-α的含量及提升血清IL-10的含量,对滑膜组织ICAM-1、VEGF、MMP3蛋白表达调控作用达到缓解滑膜炎症、减轻关节肿胀、延缓关节骨破坏的病理基础。综合来看尖叶假龙胆高剂量组优于中剂量组和低剂量组。
[Abstract]:Aim: to study the pharmacodynamics of pseudogentian on adjuvant arthritis rats.Methods: rats with adjuvant arthritis were randomly divided into three groups: model group, blank group, positive group (Tripterygium wilfordii, high dose group, middle dose group, low dose group, 8 rats in each group).In addition to the blank group, the other rats were subcutaneously injected with CFAA 0.1ml, a complete adjuvant of CFAA, to induce inflammation, and the model of AA was reproduced. The blank group was injected with normal saline in the same way.From day 14 to day 14, gavage lasted for 18 days.The weight of rats, the swelling degree of right foot and toe, the content of TNF- 伪 IL-10 in rat serum and the expression of VEGFICAM-1 MMP-3 in synovial tissue were detected by Elisa.Results the weight of AA rats was increased, the swelling of toes, arthritis score of AA rats and the formation of white pulp in spleen of AA rats were decreased.In order to alleviate adjuvant arthritis, different dose groups of Pseudogentis auriculata could reduce the content of TNF- 伪 in serum and increase the content of serum IL-10 in AA rats, so as to relieve adjuvant arthritis.The expression of VEGFU ICAM-1 MMP3 protein in synovial tissue of AA rats could be reduced in different dose groups of pseudogentian, so as to relieve synovitis, alleviate joint swelling and delay joint bone destruction.Conclusion: the apical leaf pseudogentian can effectively inhibit the secondary joint swelling, decrease the content of TNF- 伪 in serum and increase the content of serum IL-10 in AA rats.By regulating the expression of ICAM-1 VEGFN MMP3 protein in synovial tissue, the pathological basis of relieving synovitis, reducing joint swelling and delaying joint bone destruction can be achieved.In a word, the high dose group was superior to the middle dose group and the low dose group.
