四川呼吸道合胞病毒F蛋白基因序列和抗原表位变异分析

【摘要】:目的分析呼吸道合胞病毒(RSV)四川分离株F蛋白基因序列,探讨F蛋白基因和抗原表位变异情况。方法 RT-PCR方法扩增F蛋白近全长序列并测序,分析核苷酸和氨基酸变异位点,比较不同亚型和基因型间中和表位、细胞毒性T淋巴细胞(CTL)表位基因组成。结果 10株RSV F蛋白的核苷酸和氨基酸P-distance分别为0.102±0.005和0.058±0.006,中和表位47F和L4在亚型间高度保守,而RS-348和7C2在亚型内保守,CTL表位HLA B*57、HLA A*01和HLA Cw*12也是亚型内高度保守,亚型间存在个别氨基酸变异。结论 RSV四川分离株F蛋白基因保守度较高,抗原表位在亚型内保守,已在A亚型上识别的CTL表位可能在B亚型内不能识别。

Methods the near-full-length sequence of F protein was amplified and sequenced by RT-PCR. The variation sites of nucleotides and amino acids were analyzed. The neutralization epitopes among different subtypes and genotypes and the (CTL) epitope gene composition of cytotoxicity T lymphocytes were compared.
