上调miRNA-21对骨髓间充质干细胞的影响

目的探讨miRNA-21对大鼠骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)特性的影响。方法 BMSCs使用DMEM液进行培养传代,流式细胞仪检测BMSCs的表面相关抗原表达。建立miRNA-21慢病毒干扰载体(LV-miRNA-21),并转染BMSCs。分3组细胞进行对比实验[CK(正常组)、NC(对照组)、miRNA-21组],分别培养3 d、7 d和14 d;利用免疫荧光染色技术和Western-blotting技术检测并比较肌钙蛋白在3组细胞中3个时间点的表达情况,噻唑蓝(MTT)法检测3组BMSCs的增殖情况,运用流式细胞技术检测3组BMSCs的细胞周期及凋亡情况。结果 BMSCs呈长梭形不规则形态,表现出干细胞特异性抗原CD90和CD29阳性,CD45阴性。成功构建LV-miRNA-21;发现在miRNA-21组BMSCs中经诱导分化后肌钙蛋白表达量明显高于CK组和NC组(P<0.05),在miRNA-21组中表现出细胞的增殖加快,但凋亡速度比CK组和NC组减慢。结论 miRNA-21影响BMSCs的增殖和凋亡,可...
