NGF在大鼠慢性偏头痛模型中的表达及机制研究

背景:我们前期研究发现,三叉神经脊束尾核(trigeminal nucleus caudalis,TNC)中酸敏感离子通道3(acid-sensing ion channel 3,ASIC3)参与了反复发作性偏头痛的病理过程。神经生长因子(nerve growth factor,NGF)可以诱导许多疼痛疾病的痛觉敏化,并且可以调控ASIC3的表达。然而,NGF在慢性偏头痛(chronic migraine,CM)中的作用尚不清楚。所以我们探究了CM大鼠TNC部位NGF的表达,NGF对ASIC3的调控作用,及相关的信号通路在炎性汤(inflammatory soup,IS)诱导的皮肤痛觉过敏中的作用。方法:1.通过硬脑膜反复滴注7日IS建立CM大鼠模型,模拟CM的发作。von Frey测定大鼠眶周及足底的痛阈值,以此评价模型的建立。2.利用PCR,Western blot检测TNC部位NGF m RNA水平及蛋白水平,免疫荧光检测NGF在TNC神经元中的定位情况。3.CM大鼠侧脑室注射NGF中和抗体(anti-NGF neutralizing antibody,anti-NGF),von...
