AH6809干预的单纯性脑震荡大鼠海马mPGES-1的表达变化

【摘要】:目的:观察单纯性脑震荡(pure cerebral concussion,PCC)致伤后大鼠海马膜结合型前列腺素E2-合酶1(membrane-associated prostaglandin E synthase-1,mPGES-1)在EP2受体抑制剂(AH6809)干预后的表达变化,探讨AH6809对mPGES-1的干预调控作用。方法:采用金属单摆闭合式脑损伤打击装置制备PCC模型,分别于致伤后即刻和12 h腹腔注射AH6809,然后随机分为1、2、3 d和7 d组(n=5),另设相应生理盐水对照组(n=5)。采用免疫组织化学染色和Western Blot方法,观察PCC+AH6809组和生理盐水对照组大鼠海马(CA1~4和上、下齿状回)内m PGES-1的表达情况。结果:免疫组化实验结果显示:在AH6809干预前后均可观察到mPGES-1阳性产物表达于海马CA1~4和上、下齿状回的神经元胞体,呈圆形或卵圆形,胞核不着色,胞浆为棕黄色。与生理盐水对照组相比,AH6809干预后2 d组均出现mPGES-1阳性表达相对减弱,染色渐浅且细胞轮廓不清,3 d组下调最为明显,其中3 d组和7 d组的差异具有显著性(P0.05)。Western Blot实验结果显示:mPGES-1蛋白出现相似的规律性变化,下降的低峰出现在7 d组,其中2、3、7 d组与对照组相比差异具有显著性(P0.05)。结论:AH6809对PCC大鼠海马mPGES-1的表达具有一定的干预抑制作用。
[Abstract]:Aim: to observe the expression of membrane-associated prostaglandin E2-synthase (PGES-1) in the hippocampus of rats after injury induced by pure cerebral concussion.Methods: the expression of membrane-associated prostaglandin E synthase-1mPGES-1 (mPGES-1) in the hippocampus of rats after injury induced by pure cerebral concussion-PCC-1 was observed in the presence of EP2 receptor inhibitor AH6809. To investigate the effect of AH6809 on the intervention and regulation of mPGES-1. Methods: the model of PCC was established by metal pendulum closed brain injury device. AH6809 was injected intraperitoneally immediately and 12 hours after injury, and then randomly divided into 2 groups (n = 3) and 7 days (n = 7). The corresponding saline control group (n = 5) was treated with immunohistochemical staining and Western Blot method. The expression of mPGES-1 in hippocampus of PCC AH6809 group and normal saline control group was observed. Results: before and after AH6809 intervention, the positive expression of mPGES-1 was observed in CA1~4 and upper dentate gyrus of hippocampus. The neuronal bodies of the inferior dentate gyrus were round or oval, the nuclei were not stained, and the cytoplasm was brown. Compared with the normal saline control group, the positive expression of mPGES-1 was decreased relatively in the control group 2 days after intervention. The down-regulation was most obvious in the groups with light staining and unclear cell profile. The difference between the groups of 3 days and 7 days was significant (P 0.05). Western Blot results showed that the protein of 1% mPGES-1 showed similar regular changes, and the low peak of the decrease appeared in the group of 7 days. There was significant difference between the control group and the control group on the 7th day. Conclusion: AH6809 can interfere with the expression of mPGES-1 in hippocampus of PCC rats.
