燕麦β-葡聚糖对脓毒症小肠损伤相关基因表达的影响

【摘要】:目的探索燕麦β-葡聚糖对脓毒症小肠损伤相关基因表达的影响。方法 SD大鼠随机分为正常对照组(NC组),实验对照组(TC组),燕麦β-葡聚糖组(Oglu组)。TC组和Oglu组以盲肠结扎穿孔(cecal ligation and puncture,CLP)法建立脓毒症模型。Oglu组以不同剂量燕麦β-葡聚糖灌胃。12 h后收集各组小肠组织,Western blot检测小肠组织损伤基因表达。结果 1)燕麦β-葡聚糖减轻脓毒症诱导的小肠损伤,降低脓毒症大鼠小肠组织中TNF-α、IL-1β及IL-6水平(P0.05);(2)脓毒症组小肠损伤相关蛋白Bcl-2表达下调,Bax、caspase-3、Toll样、凋亡基因FAS和FASL、NF-κB表达明显增加;(3)燕麦β-葡聚糖组小肠粘膜Bax、caspase-3、Toll样、凋亡基因FAS和FASL、NF-κB的表达低于明显脓毒症组,而Bcl-2表达较高。结论燕麦β-葡聚糖调控脓毒症大鼠小肠损伤基因的表达,保护小肠上皮免受脓毒症的损伤。
[Abstract]:Objective to investigate the effect of oat 尾 -glucan on the expression of genes associated with small intestine injury in septic rats. Methods SD rats were randomly divided into normal control group (NC group), experimental control group (TC group), oat 尾 -glucan group (Oglu) group (Oglu group) and Oglu group (n = 10). Sepsis model. Oglu group was perfused with different doses of oat 尾-glucan for 12 h to collect small intestine tissue to detect the expression of small intestinal tissue damage gene by Western blot. Results 1) oat 尾-glucan alleviated the expression of small intestine tissue damage gene. 1) Oglu group was treated with different doses of oat 尾 -glucan by gastric perfusion for 12 hours to detect the expression of small intestinal tissue damage gene. Sepsis induced intestinal injury, Decreasing the levels of TNF- 伪 IL-1 尾 and IL-6 in the small intestine of sepsis rats (P0.05). The expression of small intestinal injury related protein (Bcl-2) in septic rats was down-regulated, and the expression of apoptotic genes FAS and FASLNF- 魏 B was significantly increased in the intestinal mucosa of oat 尾 -glucan group, and the expression of Bax-caspase-3 and Toll was significantly increased in the small intestine mucosa of sepsis group, and the expression of apoptotic genes FAS and FASLNF- 魏 B was significantly increased in the intestinal mucosa of oat 尾 -glucan group. The expression of apoptotic genes FAS and FASLN- 魏 B was lower than that of the patients with obvious sepsis, while the expression of Bcl-2 was higher. Conclusion Oat 尾 -glucan can regulate the expression of small intestine injury genes in septic rats and protect the intestinal epithelium from sepsis.
