Orexin-A参与迷走神经电刺激上调脑外伤昏迷大鼠前额叶皮层NMDA受体亚基NR1的表达

【摘要】:目的:探讨迷走神经电刺激(vugal nerve stimulation,VNS)对TBI后昏迷大鼠的促醒作用及其对大鼠前额叶皮层N-甲基-D-天冬氨酸(N-methyl-D-aspartate,NMDA)受体亚基NR1表达的影响,并阐明Orexin-A在其中所起的在作用。方法:将SD大鼠随机分为4组(每组36只):空白对照组(Control)、模型组(TBI)、刺激组(TBI+VNS)和orexin-1受体(OXlR)拮抗剂组(TBI+SB334867+VNS)。大鼠经“自由落体法”建立TBI昏迷模型后进行VNS治疗。根据各组大鼠的行为学变化进行意识分级,并于VNS后的6、12、24小时每组分别处死12只大鼠,取其前额叶皮层组织,应用免疫组化、Western-blot技术检测NR1表达的变化。结果:刺激组有23只苏醒,而模型组中仅苏醒11只,与此同时,刺激组大鼠前额叶皮层NR1的表达水平要明显高于模型组(P0.05)。OX1R拮抗剂组苏醒的大鼠(16只)少于刺激组,且NR1的表达也较刺激组显著降低(P0.05)。另外,模型组大鼠NR1的表达水平较空白对照组也显著降低(P0.05)。组内比较的结果显示,NR1的表达水平在模型组中6、12、24小时呈现逐渐上升的趋势(P0.05),而在刺激组中的三个时间点呈现依次递减的趋势(P0.05)。结论:迷走神经电刺激有助于脑外伤后昏迷大鼠意识状态的恢复,且能够上调大鼠前额叶皮层NR1的表达。此外,VNS促醒的作用机制可能与Orexin-A对NR1表达的调控有关。
[Abstract]:Aim: to investigate the effects of vagal nerve stimulation (VNSs) on the prowakening of coma rats after TBI and on the expression of N- methyl-Daspartate N-methyl-D-aspartate receptor subunit (NR1) in the prefrontal cortex of rats, and to elucidate the role of Orexin-A in the expression of N-methyl-Daspartate receptor subunit (NMDAR) in the prefrontal cortex of rats. Methods: SD rats were randomly divided into 4 groups (36 rats in each group: control group, model group, stimulation group) and orexin-1 receptor OXlR antagonist group. TBI coma model was established by free fall method in rats and then treated with VNS. According to the behavioral changes of rats in each group, the consciousness was graded, and 12 rats were killed in each group at 612h 24 hours after VNS. The prefrontal cortex tissues were taken and the expression of NR1 was detected by immunohistochemistry Western-blot technique. Results: 23 rats were awake in the stimulation group, but only 11 in the model group. At the same time, the expression level of NR1 in the prefrontal cortex in the stimulation group was significantly higher than that in the model group (P 0.05) .OX1R antagonist group (16 rats) was less than the stimulation group. The expression of NR1 was significantly lower than that of stimulation group (P 0.05). In addition, the expression of NR1 in the model group was significantly lower than that in the control group. The results of comparison within the group showed that the expression of NR1 in the model group showed a gradual upward trend in 24 hours, while the three time points in the stimulation group showed a decreasing trend of P0.05 in turn. Conclusion: vagal electrical stimulation is helpful to the recovery of conscious state and up-regulates the expression of NR1 in prefrontal cortex of rats after brain injury. In addition, the mechanism of VNS may be related to the regulation of NR1 expression by Orexin-A.
