多脏器功能衰竭犬室性心律失常与KCNE1关系的研究

【摘要】:目的:探讨多脏器功能衰竭(MODS)犬室性心律失常的发生与KCNE1基因的关系。方法:选择健康杂种犬14只,雌雄不限。随机分为MODS模型组7只,对照组7只。实验组失血性休克复苏后静脉注射内毒素形成"二次打击"制作犬MODS模型。当MODS犬发生室性心律失常,抢救无效死亡后,迅速取出犬的心脏,剪下犬的左心室,置于液氨中固定,后转移至-80℃冰箱中保存。提取总RNA,反转录为cDNA。采用实时荧光定量聚合酶反应(qRT-PCR)法和蛋白免疫印迹(Western blot)方法检测左心室KCNE1基因在mRNA和蛋白质水平的表达,研究MODS犬室性心律失常与KCNE1的关系。结果:与对照组相比,MODS模型组KCNE1的mRNA和蛋白质表达水平显著降低(P0.05)。结论:KCNE1与室性心律失常的发生具有相关性,其低水平表达可能是引起MODS犬发生室性心律失常的重要因素。
[Abstract]:Objective: to investigate the relationship between ventricular arrhythmia and KCNE1 gene in (MODS) dogs with multiple organ failure. Methods: 14 healthy mongrel dogs, male and female, were selected. They were randomly divided into MODS model group (n = 7) and control group (n = 7). After hemorrhagic shock resuscitation, endotoxin was injected intravenously to form a canine MODS model.
