黄体酮减轻大鼠蛛网膜下腔出血后早期脑损伤

【摘要】:目的:探讨黄体酮对蛛网膜下腔出血(subarachnoid hemorrhage,SAH)后早期脑损伤(early brain injury,EBI)中细胞凋亡、血脑屏障(blood-brain barrier,BBB)的稳定性、脑水肿和病死率保护作用。 方法:66只大鼠被随机分为假手术组,SAH+溶剂组,SAH+黄体酮组。于SAH模型制成后1、6h和12h分别给予黄体酮(16mg/kg)或等体积的溶剂。在SAH后24h分析不同组间大鼠在病死率、神经功能评分、脑水肿、细胞凋亡、caspase-3及基质金属蛋白酶-9(MMP-9)表达水平的差异。 结果:与SAH+溶剂组比较,黄体酮治疗显著降低了大鼠的病死率、细胞凋亡程度以及caspase-3水平与MMP-9的表达水平,减轻了脑水肿和伊文思蓝的渗出,提高了神经功能评分(P0.05)。 结论:黄体酮可通过抑制细胞凋亡和稳定BBB减轻SAH后EBI。
[Abstract]:Aim: to investigate the effects of progesterone on apoptosis, stability of blood-brain barrier (blood-brain barrier,BBB), cerebral edema and mortality in early brain injury (early brain injury,EBI) after subarachnoid hemorrhage (subarachnoid hemorrhage,SAH). Methods: 66 rats were randomly divided into sham-operation group, SAH solvent group and SAH progesterone group. Progesterone (16mg/kg) or the same volume of solvent was given at 1h and 12h after the SAH model was made. 24 hours after SAH, the differences of mortality, neurological function score, brain edema, apoptosis, caspase-3 and matrix metalloproteinase-9 (MMP-9) expression in different groups were analyzed. Results: compared with SAH solvent group, progesterone significantly decreased the mortality, apoptosis degree, caspase-3 level and MMP-9 expression, reduced brain edema and Evans blue exudation, and increased the neurological function score (P0.05). Conclusion: progesterone can reduce EBI. after SAH by inhibiting apoptosis and stabilizing BBB.
